汉坦病毒感染宿主细胞后TRAIL受体的动态变化

【摘要】 两株汉坦病毒(HV76/118株和H-114株)分别感染人胚肺成纤维细胞(HEPF)后,于感染后不同时间(6h、24h、96h)提取细胞总RNA,半定量RT-PCR的方法检测对肿瘤坏死因子相关诱导凋亡配体(TNF-related apoptosis-inducing ligand,TRAIL)相关受体(TRAIL-R1~R4)的表达的动态变化,以明确TRAIL受体在汉坦病毒感染细胞过程中的表达情况。结果显示,正常细胞内TRAIL-R4表达最高,其次为TRAIL-R2。汉坦病毒感染HEPF6h时仅TRAIL-R1表达显著上升,TRAIL-R2、R4表达先下降后上升并在96h达到高峰。HV76/118株感染HEPF细胞6h时TRAIL-R3表达减低并维持在低表达水平,而HV H-114感染后仅24h出现短暂下降。结论 TRAIL受体表达在汉坦病毒感染过程中呈动态变化,且不同毒株的变化趋势存在差异。

【Abstract】 To investigate the expression pattern of TRAIL (TNF-related apoptosis-inducing ligand) receptor induced by hantavirus (HV) infection,RT-PCR was employed to half-quantitate the expression of the TRAIL receptors (TRAIL-R1~R4) in human embryonic pulmonary fibroblast (HEPF) which was infected with Hantaviruses 76-118 strain and H-114 strain in 6h,24h and 96h respective.The results showed that TRAIL-R4 and R2 were found to express in normal HEPF cells.Only TRAIL-R1 was upregulated at 6h post infection.The expression of TRAIL-R2 and R4 decreased at early stage after infection,and increased at 96h post infection.The expression of TRAIL-R3 was down-regulated in H-114 infected group rather than HV H-114 group.In conclusion,the expression of TRAIL receptors show dynamic change during the process of HEPF infected with HV,and a various pattern are exhibited in different subtype HV infected groups.
