羊种布鲁氏菌菌株分子分型研究

【摘要】 目的对广东省1965-2008年分离的35株羊种布氏菌菌株进行分子分型研究。方法采用常规方法及PCR方法鉴定所分离的菌株,用脉冲场凝胶电泳(PFGE)对菌株进行分子分型分析。结果 2006-2008年分离的30株菌株均为羊种3型,种特异性M基因阳性,35株布氏菌分为14个PFGE型别,菌株间的PFGE分型相似值介于75.5-100%。结论 PFGE图谱经聚类分析显示,1965年的5株菌同源性高达100%,与近四年的菌株同源性只有75.5%,存在较大变异,表现出年度之间的差异,1965年的羊种与近四年的羊种3型的基因组存在差别,可能来自不同的克隆。PFGE作为传统生物分型的补充手段,可在种的水平对布鲁氏菌进行分型。

【Abstract】 Objective: To identify the characters and homology of brucella stains isolated in Guangdong province in the years of 2005 to 2006 and 1985. Methods: 6 brucella stains were identified by conventional methods including serological test and phage typing,along with species-specific BCSP31 gene PCR amplification.The molecular typing was performed by PFGE and the homology among those strains was analysed by using Bionumerics software. Results: The BCSP31 genes of 6 brucella strains in 2005 and 2006 were positive on genetic level and were identified as Brucella melitensis biovar 3 on phenotype level. The dendrogram shows the similarity were 69.2% to 100% among those strains. Conclusion: The species characters had changed compared with the strains in 1985. Three species of brucella can be distinguished by PFGE method.
