维汉两个民族肠道菌群与高尿酸血症关系的研究

【摘要】 目的:了解不同民族肠道微生物群落的种群分布、遗传多样性,并通过分析比较不同民族高尿酸血症患者与正常人群肠道菌群的变化规律,揭示肠道菌群与高尿酸血症的关系。方法:采用病例对照方法结合16S rDNA-PCR/DGGE技术对新疆维吾尔族和汉族高尿酸血症患者和正常人群肠道菌群进行研究,结果:两个民族人群肠道中细菌主要归12个属,高尿酸组和正常组之间差异有统计学意义(χ~2=143.415,P=0.000)。汉族和维吾尔族肠道菌群分布差异也有统计学意义(χ~2=102.534,P=0.000)。结论:虽然受到饮食结构和技术限制等方面因素的影响,本次研究通过DGGE技术发现高尿酸组和正常组以及民族之间的差异性并不是很大,但是仍然表现出一定的差异,希望在今后更为先进的检测技术的指导下对于人类肠道菌群与疾病的研究更加准确和具有科学性,为减少高尿酸血症及其他代谢性疾病的发生提供科学指导。
【Abstract】 Objectives Analysis the relationship between hyperuricemia and gut microbiota in Han and Uyger people,aim was to found the relationship of hyperuricemia and gut microbiota in different ethnic. Methods The fecal samples were collected in hospital and identification for gut microbiota by 16S rDNA-PCR and DGGE technology,then BLA ST of the sequence in GenBank indicated the species. Results we have identified 540 bacteria strains from fecal of gutThe sequence of its 16S rDNA and DGGE indicates that there is over 98%homology to the representative strains,there are 13 mainly strain in two ethnicties populations.Gut microbiota in Han was difference with Uygur ethnicity(P<0.05),still different between hyperuricemia pentients and normal people.Conclusions The mainly strain had little difference in different ethnicties and different patients because the fecal gut micribiota were influenced by diet and limited by lab technology,but still we found some diference in diferent ethnic,we hope use moden and effective technology to found the relationship between gut microbiota and diease in future,give scientical guide for treat and control hyperuricemia.
