Effect of Paeoniflorin on TLR2 mediated the inflammatory response signal transduction pathways

【摘要】 <正>Objective To investigate the effects of paeoniflorin(Pae) on the TLR2 signaling pathway in human THP-1 cell line stimulated by BLP.Methods Pae at various concentrations incubated with BLP stimulated THP-1 cells.TNF-a and IL-6 were detected using available ELISA kit.Pae incubated with BLP stimulated THP-1 cells.Western blot was used to test the expression of NF-k B in nuclear and TLR2, MyD88 in cytoplasm.The impact of Pae and BLP on cell apoptosis of THP-1 cells was estimated by flow cytometry.Results Pae dose-dependently inhibited the levels of TNF-a and IL-6 that were increased by BLP stimulation.Treatment with BLP led to an activation of NF-_κB p65 and this activation was inhibited by Pae,whereas no
