锌影响镉对体外诱导未成熟人树突状细胞的毒性作用

【摘要】 目的探讨锌对镉化合物诱导人未成熟树突状细胞(iDC)毒性作用的影响。方法分离人外周血单核细胞,体外诱导获得iDC;分别给予生理盐水(对照组),氯化镉(CdCl2组);硫酸锌(ZnSO4组);硫酸锌预处理后给予氯化镉(Zn+Cd组);锌离子特异性螯合剂TPEN预处理后给予氯化镉(TPEN+Cd组)处理。用噻唑蓝颜色反应法(MTT)检测细胞活力;单细胞凝胶电泳(SCGE)检测细胞DNA单/双链断裂损伤(SSB/DSB);吖啶橙(AO)与碘化丙啶(PI)双染色法检测细胞凋亡。结果与对照组相比,iDC活力随CdCl2剂量增加明显下降,差别有统计学意义(p<0.05);CdCl2、Zn+Cd、TPEN+Cd三组与ZnSO4组相比,在相同CdCl2剂量时细胞活性均明显下降(p<0.05);相同CdCl2剂量(20μM)下,Zn+Cd和TPEN+Cd组细胞DNA损伤程度(尾长和尾矩)均明显增高(p<0.05);各组细胞凋亡情况存在差别。结论镉化合物对诱导的人iDC有细胞毒性,且高于锌化合物;给予iDC硫酸锌(20μM)干预或耗竭细胞内锌离子增加氯化镉诱导iDC细胞DNA断裂损伤相关毒性的敏感性。
【Abstract】 Objective To study the effects of zinc compounds on the cytotoxicity of cadmium chloride to human immature dendritic cells(iDC) in vitro.Methods Cells were subgrouped: control(normal saline,NC);zinc supplementation(pretreatment with zinc sulfate 12h before cadmium administration,Zn+Cd);zinc depletion(pretreatment with TPEN 4h before cadmium administration,TPEN+Cd);cadmium treatment(treated with varied concentration of cadmium chloride,CdCl2);zinc treatment(treated with varied concentration of zinc sulfate,ZnSO4).Cell viability was conducted by MTT assay;DNA brands breaks were detected with alkaline comet assay;cell apoptosis was observed through acridine orange(AO) and propidine iodide(PI) dyeing. Results Compared with NC,cell viability decreased significantly in a dose-response manner (p<0.05);CdCl2,Zn+Cd and TPEN+Cd treatment exerted greater effects on cell viability than ZnSO4 did,at the same concentration(5、10、20、40、80、160μM).DNA damage and cell apoptosis in Zn+Cd and TPEN+Cd group was much severe(p<0.05) and aggravated than 20μM CdCl2 respectively.Conclution The cytotoxicity of cadmium chloride to iDC is stronger than zinc sulfate. Zinc supplementation(20μM) and depletion enhanced the cytotoxicity of cadmium chloride.
