【摘要】 目的建立小鼠急性酒精性肝损伤模型并筛选敏感检测指标。方法实验分8个组,实验组小鼠采用一次经口灌胃50%的乙醇(12ml/kg)后禁食4h、6h、8h、10h、12h、16 h、20h和24h,相应对照组灌胃给予纯净水并禁食同等时间。血液生化指标检测包括血中甘油三酯(TG)、胆固醇(CHOL)、谷丙转氨酶(ALT)、谷草转氨酶(AST)、碱性磷酸酶(AKP)、胆红素(TBIL)和Y-谷氨酰转酞酶(Y-GGT);16h实验组和对照组血中高密度脂蛋白(HDL)和低密度脂蛋白(LDL)含量;并取动物肝脏,冰冻切片苏丹Ⅲ染色后,观察肝组织脂肪变性程度。结果经口一次灌胃给予小鼠50%的乙醇后禁食16h,实验组与对照组比较呈现明显的脂质代谢紊乱,包括血中TG含量显著升高、HDL值显著降低、病理肝组织脂肪变性程度明显加重。结论 50%的乙醇经口灌胃一次后禁食16h,可造成小鼠急性酒精性肝损伤模型,可通过测定血中TG、HDL含量及肝组织脂肪变性程度等指标进行模型评价。更多还原

【Abstract】 Objective To establish the acute liver injury models in mice induced by alcohol and screen the sensitive index.Methods The mice were random divided into eight groups.The test groups were given 50%alcohol by gavage one time and executed respectively after fasting 4,6,8,10,12,16,20 and 24hours.The control groups were given the purified water and fasting identical hours.The blood biochemistry index included triglyceride,cholesterol,glutamic-pyruvic transaminase, glutamic-oxal(o)acetic transaminase,alkaline phosphatase,bilirubin and Y-glutamyl transferase.High density lipoprotein and low density lipoprotein were tested in the fasting 16 hours group.Livers were observed by pathologically with frost slice technique and SudanⅢstaining.Results The test group were giving 50%alcohol and fasting 16 hours showed the lipid metabolic disorder,including the indexes of triglyceride and high density lipoprotein significantly different from the control group(P<0.05).The fatty degeneration of me hepatic cells aggravated in the test gourp(P<0.05).Conclusion Giving 50%alcohol and fasting 16 hours could induce the acute liver injury models in mice,by testing triglyceride and high density lipoprotein in blood and the fatty degeneration of the hepatic cells.更多还原
