LC-MS/MS筛选食物中毒中天然生物碱毒素

【摘要】 目的建立亲水液相色谱一串联质谱法对食物中毒中常见的7种植物性有毒生物碱成分进行筛选分析。方法原料液液提取后,用液相色谱一串联质谱仪以电喷雾电离,多反应监浏(MRM)方式进行分析。结果应用亲水液相色谱一串联质谱法,以化合物的保留时间、两对母离子/子离子对定性,方法对7种生物碱的分析具有良好的线性响应(r>0.998),相对标准偏差低于10%。结论该方法选择性佳、灵教度高,适用于食物中毒分析鉴定。
【Abstract】 Objective Hydrophilic interaction liquid-chromatography(HILIC) coupled with electrospray tandem mass spectrometry has been developed to the separation of seven toxic alkaloids (aconitine,hypaconitine,ephedrine,gelsemine,raceanisodamine,strychnine,brucine).Method Isocratic separation and analysis were achieved for seven alkaloids using a mobile phase consisting of ammonium formate and acetonitrile at pH 3.Two MRM transitions for each substance were monitored to provide sufficient identification of alkaloid.Results Good linearity of the calibration curves were obtained (r>0.998).High reproducibity of the method was observed,with a relative standard deviation below 10%.Chromatographic conditions including percentage of organic modifier in the mobile phase,pH,and concentration of buffer salt were investigated.Conclusion The validated method enables selective identification as well as accurate and sensitive quantification and can be applied in poison case.
