SRAP和SSR标记在枇杷种质资源遗传多样性研究中的应用

【摘要】 本实验利用SRAP和SSR两种分子标记共计50对引物组合对来自6个国家的46份枇杷品种资源的基因组DNA进行了遗传多样性分析,两种标记分别获得13435和685条扩增谱带,其中434个SRAP标记位点中有236个位点产生多态性,10对SSR引物由8对产生多态性,分别利用NTSYSpc-2.10和DPS3.01软件对46份枇杷资源进行聚类分析,获得的两个聚类图结果较为吻合,仅个别资源有拉近或拉远的趋势。利用SRAP和SSR两种标记的扩增谱带数据进行聚类分析,可将46份枇杷品种划分为4个类群,并依相似性系数分析了各品种资源的亲缘关系。利用POPGENE 1.31软件对46份种质遗传多样性计算获得SRAP+SSR(469个位点)的观测等位基因数(Na)=1.592、有效等位基因数(Ne)=1.290、Nei’s基因多样性=0.174、总基因多样性Ht=0.183、群体内基因多样性Hs=0.057、平均基因分化系数Gst=0.691,说明69.1%的遗传变异来自群体间,30.9%的遗传变异来自群体内,基因流Nm=0.224,说明群体间的基因交流较少,生长在同一地区的枇杷品种遗传基础较为相似,而且比较稳定;枇杷不同地区的遗传距离变异幅度为0.015-0.271,说明来源于不同栽培区的枇杷品种间的亲缘关系较远。进一步印证了枇杷资源按照地理分布进行分类的合理性。

【Abstract】 To investigate the genetic diversity in the cultivated loquat germplasm and to provide information for parental selection and germplasm improvement,forty-six loquat germplasm from six countries were analysed with SRAP and SSR markers.The results showed that:A total of 13435 SRAP bands and 685 SSR bands were obtained with 50 informative primer combinations,236 bands among 434 SRAP bands(54.37%) and 8 of 10 SSR primer combinations were polymorphic.The clustered result using NTSYSpc-2.10 and DPS3.01 softwares showed no difference for 46 materials,only some germplasm had difference.The 46 materials were clustered into four groups according to the band of SRAP and SSR.The genetic relationship of 46 loquat cultivars was analyzed using the similarity coefficient(GS).The average value using NTSYSpc-2.10 software of observed number of alleles (Na),effective number of alleles(Ne),Nei’s gene diversity(H) and Shannon information index(I) were 1.592, 1.290,0.174 and 0.183,respectively.The mean coefficient of gene differentiation(Gst) among populations was 0.691,30.9%of which was within populations and 69.1%was among populations.The mean estimate of gene flow was 0.224,indicating that the gene flow between populations was less and the hereditary basis among the materials from same planting zone was similar and steady.The genetic distance based on the Nei and Li’s genetic similarity (GS) coefficients values among the materials from different planting zones was ranging from 0.015 to 0.271. Genetic relationship of loquat cultivars from different planting zones is distant.Cluster analysis proved the rationality of classification according to the loquat germplasm geographical distribution.
