深海来源Bacillus anmyloliquefacines SH-B10产生抗真菌脂肽的分离纯化及结构鉴定

【摘要】 对一株分离自中国南海深海3601m处海泥的解淀粉芽孢杆菌(Bacillus amyloliquefaciens)SH-B10发酵产生的抗植物病原真菌物质进行了分离纯化及结构鉴定。以黄瓜枯萎病菌尖孢镰刀菌黄瓜专化型(Fusarium.oxysporumf.sp.cucumerinum)为靶菌对抗真菌活性物质进行追踪,采用酸沉淀、快速色谱及半制备高效液相色谱(HPLC)等手段从该菌株的发酵液中分离纯化得到4个化合物。经Q-TOF-MS串联质谱技术将其中2个化合物鉴定为C16fengycin A(2)及C17 fengycin A(3)活性测试结果表明,二者对黄瓜枯萎病菌尖孢镰刀菌黄瓜专化型(F.oxysporumf.sp.cucumerinum)均具有明显抑制作用。
【Abstract】 4 antifungal lipopeptides were isolated from a deep sea derived bacterium Bacillus amyloliquefaciens SH-B10 by bioactivity guided fraction and purification using acid precipitation,flash chromatography and semi-preparative HPLC.Structure eucidation of compound 2 and compound 3 was accomplished by interpretation of Q-TOF-MS spectra. Compound 2 and 3 showed significant inhibitory acitivty against plant pathogen fungus Fusarium oxysporum f.sp.cucumerinum.
