应用变性高效液相色谱快速检测鉴定花生栽培品种的研究

【摘要】 研究构建了基于变性高效液相色谱(DHPLC)的花生指纹图谱,建立了16种花生栽培品种快速检测鉴定技术。以16个花生品种的DNA为检测目标,选取3组花生微卫星DNA简单重复序列(SSR)引物对进行降落PCR(TouchDownPCR)扩增,产物分别采用琼脂糖凝胶电泳和DHPLC进行检测。结果表明,无论采用3组引物中任何一种,琼脂糖凝胶电泳均无法准确区分16种花生品种,而在DHPLC检测中,当采用引物组L时,其所建立的DHPLC指纹图谱能够准确鉴定全部16种待测样品。这表明SSR-PCR-DHPLC技术能够成为一种快速、准确的花生栽培品种检测鉴定技术。

【Abstract】 The fingerprinting map of peanut was established by denaturied high performance liquid (DHPLC) and the rapid determination and identification methods of 16 cultivars were developed.The DNAs of 16 cultivars were amplified by TD PCR(Touch down PCR) with 3 selected groups of SSR(Simple sequence repeats) markers primer pairs,then the amplicatic products were detected by agrose gel electrophoresis and DHPLC.The peanut cultivars could not be distinguished each with of 3 selected groups by agrose gel electrophoresis,while 16 cultivars could be distinguished with primer group L by DHPLC fingerprinting map.SSR-PCR-DHPLC is a specific,sensitive,rapid and highly effective method for the detection and identification of peanut cultivars.
