微流控芯片非接触电导法分离检测盐酸普萘洛尔与酒石酸美托洛尔
Separation and Determination of Propranolol Hydrochloride and Metoprolol Tartrate by Microfluidic Chip with Contactless Conductivity Detection

【摘要】 建立了微流控芯片非接触电导法快速分离检测盐酸普萘洛尔和酒石酸美托洛尔的分析方法。考察了电泳介质的种类、浓度、pH值、添加剂种类及用量、对照品与样品的溶剂、进样时间、分离电压等因素对分离检测的影响。结果表明,选择7 mmol·L-1 Tris+21 mmol·L-1H3BO3(pH=7.5),进样时间为15.0 s,分离电压为2.0 kV时可实现较好的分离和检测。盐酸普萘洛尔和酒石酸美托洛尔的线性范围分别为4.0×101~1.2×103mg·L-1和3.0×101~1.0×103mg·L-1,相关系数分别为0.997、0.998;检出限(S/N=3)分别为10、20 mg·L-1;加标回收率分别为98%、105%,平均RSD(n=6)为1.2%、1.1%。该方法快速、简便、灵敏,可用于β-受体阻断类药物的质量控制。
【Abstract】 A method for determination of propranolol hydrochloride and metoprolol tartrate by micro-fluidic chip with contactless conductivity detection was established.The electrophoretic parameters, such as the variety and concentration of buffer solution,separation voltage etc.were researched.Propranolol hydrochloride and metoprolol tartrate were separated and detected in buffer of 7 mmol ? L-1 Tris + 21 mmol ? L-1 H3BO3 at 2.0 kV of constant voltage.The linear concentrations of propranolol hydrochloride and metoprolol tartrate ranged from 4.0×10 to 1.2×103 mg ? L-1(r =0.997),from 3.0×101 to 1.0×103 mg ? L-1(r =0.998) respectively,the limits of detection(LOD) reached 10 mg ? L-1,20 mg ? L-1(S/N = 3 ),respectively.The average recoveries were 98%and 105%respectively. The average RSDs(n.=6) were 1.2%and 1.1%,respectively.This study provides an effective and simple method for the quality control of the product.
