两种德国鸢尾的组织培养技术

【摘要】 以两种德国鸢尾‘金娃娃’、‘黑骑士’的幼嫩花莛为材料,研究不同外植体、不同消毒方法和不同激素配比对2种鸢尾不定芽诱导、不定芽增殖、生根的影响。结果表明:两种鸢尾的花莛仅花茎上苞片着生处的结节能诱导出不定芽;‘金娃娃’外植体以75%乙醇(体积分数)浸摇3s、0.1%HgCl2(质量分数)消毒6min污染率较低,存活率较高;‘黑骑士’则以75%乙醇浸摇5s、0.1%HgCl2消毒8min效果较好;初代培养中,Ms+2.0mg/L 6-BA+0.1mg/L NAA是2种鸢尾不定芽诱导的最佳培养基。其中对‘金娃娃’的不定芽诱导率为86.4%,对‘黑骑士’的不定芽诱导率为73.5%,是不定芽诱导的最佳培养基;继代培养中,MS+4.0mg/L 6.BA+0.2mg/L NAA是继代培养的最佳培养基,对‘金娃娃’的增殖系数为12.71,对‘黑骑士’的增殖系数为8.64,是继代培养的最佳培养基;生根培养以MS+0.1mg/L 6-BA+2.0mg/L NAA效果最好。
【Abstract】 Young scapes Iris germanica ’ Gold Boy’ and ’ Black Knight’ were used as materials,different explants,disinfection methods and plant hormones combinations of medium on tissue culture were studied to discuss the induction of adventitious buds,proliferation of adventitious buds,rooting of buds of two irises.The results showed that only the tubers of flower stalks where the flower bracts grew of both two irises can adventitious buds be induced;the disinfection of soaking for 3 seconds in 75%alcohol(volume fraction),6 minutes in O.1%HgCl2(mass fraction) was low in both contamination rate and death rate for Iris germanica ’Gold Boy’,while for ’Black Knight’,5 seconds in 75%alcohol,8 minutes in 0.1%HgCl2,instead;The optimal medium of primary culture for the induction of adventitious buds was MS + 2.0 mg/L 6-BA + O.1mg/L NAA,whose induction rates were 86.4%for ’Gold Boy’ and 73.5%for ’Black Knight’;As for secondary culture,MS+4.Omg/L 6-BA + O.2mg/L NAA was the optimal medium with the highest proliferation rates of 12.71 for ’ Gold Boy’ and 8.64 for ’ Black Knight’;MS+0.1 mg/L 6-BA+2.0 mg/L NAA was optimal for rooting.
