响应面法优化南瓜籽中不皂化物提取的皂化工艺条件

【摘要】 南瓜籽不皂化物中含有植物甾醇、角鲨烯和生育酚等活性成分,是潜在的功能性食品、化妆品、药物的营养成分。本文以不皂化物的质量为指标,采用响应面法优化了南瓜籽中不皂化物提取的皂化工艺条件,为南瓜籽的开发利用提供理论参数。利用单因素试验考察了皂化时间、皂化温度、液料比、KOH-乙醇浓度对不皂化物提取率的影响,在此基础上采用响应面法(RSM)中的Box-Behnken设计试验对皂化工艺进行优化。结果表明,在皂化时间为1.5h、皂化温度为69℃、液料比为9.7:1(mL/g)和KOH-乙醇溶液浓度为2.06mol/L不皂化物提取率为1.3702%,误差为0.44%,与理论值基本相符。说明采用Box-Behnken试验设计寻求的最佳皂化工艺条件可行。

【Abstract】 Unsaponifiable matter from Pumpkin seed contain many bioactive compounds,such as phyotosterol,squalene and tocopherol,which potential as functional food,cosmetics, pharmaceutical ingredient.In the present study,based on the mass of unsaponifiable matter, saponifying extraction of unsaponifiable matter from Pumpkin seed was studied by adopting response surface methodology(RSM),aiming to provide theoretical parameters for its further exploitation.Four factors of reaction time.temperature,liquid / material ratio and KOH-ethanol concentration were investigated on the basis of single factor test,by Box-Behnken design and analyzing the response surface plots,the extraction parameters were optimized.Results showed that the optimum parameters for saponification were saponification time was 1.5h,saponification temperature is 69℃,the rate of liquid to material ratio was 9.7:1(mL/g) and KOH-ethanol concentration was 2.06mol/L,the yield of unsaponifiable was 1.3702%,with the error of o.44%, which was close to the theoretic value,thus,Box-Behnken design is a good way to seek the optimal saponifying extraction conditions.
