结球甘蓝BoTCTP基因表达载体的构建及转化芥蓝的研究

【摘要】 芥蓝(Brassica oleracea var.alboglabra Bailey)是十字花科重要蔬菜之一,其风味别致、营养价值高,深受消费者欢迎。但是,在芥蓝的栽培过程中常常会受到各种逆境的侵害,给生产造成很大损失。翻译控制肿瘤蛋白(TCTP)的功能主要有促进细胞增殖和植物生长、抗高低温、盐碱、重金属等逆境条件的作用。本研究根据结球甘蓝翻译控制肿瘤蛋白基因(BoTCTP)序列设计特异引物,经PCR扩增得到目的片段,然后构建表达载体,利用农杆菌介导法导入芥蓝中,得到了转基因植株,并对其进行了分子生物学鉴定,最终得到了4株转基因植株,其外源目的基因已经整合到受体基因组中,1株以双拷贝形式存在,另外3株以单拷贝形式存在。阳性植株相对于阴性对照生长加快,叶片面积较大、株高明显高于阴性对照等。在高低温、盐碱、干旱、重金属等逆境胁迫下,转基因植株对胁迫的忍受力明显优于对照植株。

【Abstract】 Chinese kale(Brassica oleracea var.alboglabra) is one of the most important vegetables of Brassica oleracea.However,in Chinese kale cultivation process,the damage of various stress usually leads to great loss of its production.The function of translationally controlled tumor protein(TCTP) in plant mainly includes promoting the cell proliferation and resistance to stress such as high temperature,low temperature,salt,heavy metal and so on.In this research,the specific primers were designed according to BoTCTP.The interest fragment was amplified by PCR, and then the expression vector was constructed with it and transferred into Chinese kale mediated-Agrobacterium.The transformants were confirmed by molecular methods and biological assay.Four transgenic plants were obtained.They were taken to design the experiments associated with growth and stress such as high temperature,low temperature,salt,heavy metal and so on.The results showed that the transgenic plants grew faster,and had bigger leaves than the non-transgenic plants.In various stress condition,the transgenic plants had obviously stronger resistance than the control.
