文心兰花芽的解剖结构及组织化学研究

【摘要】 采用石蜡切片法观察了文心兰‘Milliongolds’花芽的形态发生和结构发育过程,用石蜡切片高碘酸席夫反应法(PAS法)和汞溴酚蓝组织化学染色法研究糖和蛋白质在文心兰花芽分化过程中的变化。研究表明:文心兰花芽的分化进程可以分为未分化期、花蕾原基分化期、萼片原基分化期、花瓣原基分化期、合蕊柱及花粉块分化期6个时期。糖和蛋白质主要以颗粒状分布在各器官的原基顶端及其周围。在花序原基分化期,细胞中糖的含量较少,而蛋白质含量丰富且整体分布均匀。随着花芽的继续分化,糖和蛋白质含量在花萼原基分化期最为丰富,在花瓣原基时期含量明显减少。进入合蕊柱及花粉块分化期时,糖和蛋白质在合蕊柱及花粉块着生处最为集中。
【Abstract】 The flower bud differentiation process of Oncidium‘Milliongolds’ was studied by the method of paraffin cut The results showed that flower bud differentiation process could be divided into six phases:undifferentiation phase,inflorescence priomdium differentiation phase,flower primordium differentiation phase,sepal primordium differentiation phase,petal primordium differentiation phase,and column and pollinia differentiation phase.The results showed that the saccharide and protein were mainly in or around the primordium.In inflorescence primordium differentiation phase,the content of saccharide was little, while protein was rich and distributed uniformly.With the continued differentiation of the flower bud,saccharide and protein occurred abundantly in sepal primordium differentiation phase,and they decreased as a visible grads in petal primordium differentiation phase.The saccharide and protein focused on column and pollinia during column and pollinia differentiation phase.
