甘露醇对蝴蝶兰种质离体保存的影响

【摘要】 以蝴蝶兰无菌苗为离体保存材料,研究了培养基中添加不同浓度甘露醇对蝴蝶兰试管苗保存的影响。结果表明:在培养温度24±3℃,保存前3个月光照强度2000~2500lx,3个月后光照强度500~1000lx,光照时间12 h·d-1的培养条件下,花宝1号(Hyponex No.1)3.0g·L-1+AC 0.5g·L-1+蔗糖20g·L-1培养基中添加15g·L-1甘露醇能使蝴蝶兰试管苗连续保存18个月,存活率达87.5%,比对照存活率68.8%,提高了18.7%;保存18个月的试管苗在增殖培养基上能恢复生长,与正常生长的继代苗相比,形态上没有发生变异。
【Abstract】 In vitro preservation of sterile Phalaenopsis amabilis plantlets with the addition of mannitol in the culture medium was investigated.The results showed that Hyponex No.1 3.0g·L-1+AC 0.5g?L-1+sugar 20?L-1 in the medium supplemented with 15g?L-1 mannitol could prolong plantlet’s shelf life to 18 month at(24±3)℃under the 12h photoperiod culture condition of 2000-25001x in the first 3 months,and 500-1000 lx during the next 3 months.After 18 months in the medium,the plantlet’s survival rate was 87.5%,and survival rate of 68.8%compared with the control,increased 18.7%, and the plantlet’s growth was normal after cultured in a recovering medium.
