【摘要】 本文以东方百合杂交系切花品种‘索邦’（‘Sorbonne’）为材料,在以鳞片为外植体进行启动培养的过程中,对不同种类和浓度的灭菌剂、不同灭菌剂组合方式以及不同部位鳞片的灭菌效果;不同部位的鳞片、同一鳞片的不同部位、不同的接种方式、培养基中不同的激素浓度及其配比对外植体诱导分化率上的差异等进行了科学的统计和对比分析,研究以上因素对百合鳞片启动培养的影响。实验结果表明在较长时间酒精和升汞的处理下,鳞片的灭菌效果较好。在相同的灭菌条件下,内层鳞片的污染率明显低于中层鳞片。在外植体的选择上,内层鳞片基部具有较高的诱导分化率,是最佳的外植体来源。由于几乎全部的芽点都在鳞片的近轴面上形成,所以以鳞片近轴面向上平放于培养基上的接种方式,更容易获得大量健壮的小苗。诱导不定芽的适宜培养基为MS+6-BA2.0mg·L^-1+NAA0.2mg·L^-1。更多还原

【Abstract】 In induction phase of tissue culture of scales in vitro,the results of different sterilization methods and the relations between the result of induction and scales parts of explants and hormone combination in the medium of initiation period were studied with Sorbonne’,one cultivar of Oriental Lily.The results indicated that with longer soaking time in alcohol and HgCl₂, scales have better sterilization result The rate of infection of inner layer scales was far lower than middle layer scales’ which were sterilized with the same method.The rate of induction of the base of inner layer scales was highest As almost all of shoots were formed on inner side of scales,putting explants on the medium with their outer side can obtain better induction result Better induction medium of shoots is MS +6-BA2.0mg·L^-1 + NAA0.2mg·L^-1.更多还原
