不同种百合亲本与杂交后代DNA提取方法的优化及RAPD分析

【摘要】 以山丹(Lilium.pumilum)、渥丹(Lilium.concolor)为父本,以‘白天使’(‘Navona’)为母本进行杂交,对杂交胚进行抢救获得杂交后代,并将杂交后代进行继代组织培养。提取亲本及杂交后代的总DNA,再选用不同引物,利用RAPD技术对亲本及杂交后代进行分析,判断杂交后代成功与否。研究不同条件对RAPD效果的影响,寻求最佳的分析方案。初步研究结果:选用的五种引物都能扩增出条带,比较亲本和其杂交后代的带谱看出:山丹与‘白天使’的杂交后代为真杂种;而渥丹与‘白天使’的杂交后代不能判断是否为真杂种。

【Abstract】 The hybrid progenies of Lilium pumilum×’navona’ and Lilium.concolor×’navona’ were used in this study.We did subculture on the hybrid progenies,then extracted the DNA of them and their parents.We choosed different random primers to enlarge those materials and concluded that if it was successful by RAPD analyzing to the enlarged product.At the same time we studied the impact of different conditions to the effect of RAPD analyze.The results were as follows:all primers could enlarge out different DNA string in the hybrid progenies and their parents.The hybrid progenie of Lilium.pumilum×’navona’ was successful and the other hybrid progenie was unsuccessful by comparing the DNA string of samples.
