南瓜属三个种的亲缘关系与品种的分子鉴定研究

【摘要】 用随机扩增多态DNA(RAPD)技术,对南瓜属(Cucurbita)3个主要栽培种即美洲南瓜(C.pepo L.)、中国南瓜(C.moschata D.)和印度南瓜(C.maxima D.)的23个品种(系)及种间杂交后代进行亲缘关系与品种(系)的分子鉴定研究。从200个随机引物(10bp)中筛选出17个引物,对23个南瓜品种(系)的染色组DNA进行PCR扩增,共产生80条扩增带,其中77条在南瓜属3个种间表现差异,可作为遗传标记(RAPD标记)。RAPD技术对3个基因组的分析结果与传统分类学的结果完全相符,同时RAPD技术对南瓜属内不同品种(系)间的亲缘关系的分析结果与其他地域来源和形态特征基本符合。大部分南瓜品种(系)都有其特有的扩增带可与其他品种(系)区别开来,这种特异的条带可作为一个品种的分子标记应用于品种鉴定和分类。

【Abstract】 Twenty-three cultivars from three mainly cultivated species(C.pepo L.,C. moschata D.and C.maxima D.) in Cucurbita were tested with RAPD(Random amplified polymorphic DNA) using 17 arbitrary 10-mer primer which were screened from 200 primer.The level of polymorphism within cultivars was very high.Among a total of 80 bands observed,77 (96.25%) bands were polymorphic in the 23 cultivars examined.These markers were enough to distinguish each of the species used.Genetic distances between each of the cultivars were calculated from simple matching coefficients.A dendrogram showing genetic relationships between them was conducted by an unweighted pair-group method with arithmetical averages (UPGMA).This conclusion is completely consistent with the traditional cultivar classification and fundamentally identical with their origins and characteristics.Most of the cultivars had their specific bands which were available to identifying different cultivars in the genus Cucurbita.
