基于毛细管电泳的香蕉AFLP的分子标记研究

【摘要】 本试验利用毛细管电泳技术对香蕉品种资源进行AFLP分子标记研究,在前期研究的基础上,选取两对引物EcoR ACC+Mse CAT和EcoR ACC+Mse CAG对供试材料进行了基因组水平上的多态性检测,获得了比较清晰的毛细管电泳图谱,供试的73份香蕉种质显示出丰富的多态性,共得到扩增位点245个,其中多态性位点199个,多态性位点比例达到81.2%,对73份材料的区分率达到100%。根据毛细管电泳图谱聚类分析,在相似系数0.76的水平上,将可73份香种质划分为6个类群,传统的Simmonds分类系统中的各大类型基本能聚类在一起,但其中部分品种的分类地位与Simmonds分类系统存在差异,研究结果显示粉蕉与大蕉两个类群之间遗传距离差异明显,将其简单的归为ABB类型不能反映品种间的亲缘关系。同时,对一些特殊种质进行了鉴定,结果表明儿内亚和苹果两份种质材料实际上为同一个品种。
【Abstract】 Investigation of the genetic diversiry of Banana and plantain is significate in utilizing its germplasm resources and breeding new cultivars.The phylogenetic relationships among 73 germplasm resources of Banana and plantain were analyzed by using of AFLP technique to study the genetic diversity with two AFLP primer pairs EcoRACC+MseCAT,and EcoRACC+MseCAG.Among the 245 bands detected, 199 bands were polymorphic.the high ratio of polymorphic bands(81.2%) observed indicated abundant genetic diversity among Banana and plantain.The results suggested that the two pairs of primer combinations could be used to distinguish all tested cultivars.Based on the cluster analysis of analyzed AFLP finger printing,the 73 materials of banana and plantain can be divided into 6 groups when Dice similar coefficient is 0.76.Dice similar coefficients of two samples(C41,C42)are0.98.That is to say the two samples are the same; there was very significant difference between Fenjiao and Dajiao.
