几丁质酶基因转化巴西香蕉的研究

【摘要】 为建立高效的香蕉遗传转化体系并将几丁质酶基因转到香蕉主栽品种巴西蕉中,以获得抗香蕉枯萎病的新种质,本研究以巴西蕉球茎横切薄片为转化受体,摸索了卡那霉素和G-418的最佳筛选浓度,优化了共培和筛选等条件,建立了以农杆菌介导的巴西蕉薄片遗传转化体系。试验结果表明,G-418更适合于香蕉薄片为受体的筛选用抗生素,其最佳筛选浓度50mg/L:共培则以固体-液体-固体培养基共培6天效果最好,经过对抗性芽2~3次继代筛选和PCR鉴定,试验获得5个转基因株系。
【Abstract】 In order to establish a high efficient Agrobacterium-mediated genetic transformation system for banana,and introduced chitinase gene into banana(Musa AAA Cavendish cv.Baxi) to obtain possible transformants resistance to Fusarium wilt disease.In this study,we explored the effects of different concentration of 2 antibiotics,optimized the co-cultivation and selection condition for thin-cell-layer regeneration and transgenic system,The results indicated that G-418 is more sensitivety than Kanamycin,the suitable concentration of G-418 on regeneration of shoots cultivation is 50 mg/L,the best method for co-cultivation is three-phase co-cultivation,that is semi-solid/ liquid/ semi-solid medium cultivation and co-cultured for 7 days.After 2~3 times of subculture in selection medium and identified by PCR,5 transgenic plants out of 8 regenerants were obtained.
