香蕉一个内切几丁质酶基因的表达及其与乙烯和果实成熟的相互关系

【摘要】 从SSH文库中分离到了一个内切儿丁质酶基因,该基因主要在果实和花中大量表达,而在根、茎、叶中不表达或表达量很低。对在乙烯利催熟处理、1-MCP抑制处理以及对照条件下的香蕉果实采后不同时期分别进行了该基因的差异表达分析利儿丁质酶活性测定。结果表明:乙烯利处理的香蕉果实,内切儿丁质酶基因表达量增大,表达高峰提前,这表明该基因的表达受外源乙烯的诱导,与果实成熟棚关。我们还发现,正常成熟的果实和乙烯利处理的香蕉果实,果皮中儿丁质酶的变化早且变化量远大于果肉。这说明在香蕉果实采后成熟过程中,果皮中的内切儿丁质酶起着非常重要的作用。

【Abstract】 An endochitinase gene had been isolated from banana fruit Suppression Subtractive Hybridization(SSH) library.It was mainly expressed in fruit and flower.Differential expression and chitinase activities had been investigated in different stages ofpostharvested banana fruits with or without ethylene and 1-methylcycle -propene(1-MCP) treatments.The results showed that under ethylene treatment,gene expression and chitinase activities were stronger than that of with 1-MCP treatment and the control.Moreover, with ethylene treatment,gene expression peak appeared earlier than the control and l-MCP treatment.It was clearly indicated that this endochitinase gene responsed to exogenous ethylene and was closely related to banana fruit ripening.Furthermore,the change of chitinase activities in peel was earlier and stronger than in pulp for ethylene treatment and the control,which was suggested that chitinase in peel play important role during the progress of banana fruit ripening.
