湖南宽皮柑橘和柚及近缘种的SRAP分析(摘要)

【摘要】 采用SRAP分子标记方法,对25份宽皮柑橘品种、32份柚及近缘种进行了遗传相似性分析,目的是为宽皮柑橘、柚及近缘种育种亲本合理选配和及其种质的充分发掘利用提供科学依据。结果表明:31对SRAP引物共检测到宽皮柑橘品种175个等位基因,平均每对引物检测到5.6452个,Me10-Em11和Me5-Em11的等位基因数最多,有11个,Me8-Em11最少,仅有2个;遗传相似性系数变异范围为0.58~0.94;基于SRAP标记,在相似系数为为0.785水平上,25份种质分成9大类:(1)道县光皮大果、臭皮柑;(2)滑皮橘、茶籽柑、江津酸橘、印度酸橘、砂糖橘、椪柑260、地中海柑橘、安化红早熟蜜柑、南橘、立花橘;(3)尖叶型莽山单株、本地早、南丰蜜橘、细皮狗屎柑、南柑20、福桔、怀化宜吕橙;(4)粗柠檬;(5)枸橼;(6)滑皮柑;(7)沙田柚;(8)金柑;(9)印度野橘。相似系数0.68为临界值时,可将32份柚及近缘种试材分为4个组群,沙田柚系列,文旦柚为中心并包含酸橙近缘系列,甜橙类及野生资源。SRAP标记基本能区分各个品种(系),各类均有其特征谱带。获得了湖南宽皮柑橘品种、柚及近缘种的亲缘关系。
【Abstract】 Genetic diversity of 25 loose skin mandarin landraces were analysed with Sequence related amplified polymorphism(SRAP).The aim was to provide genetic basis for breeding of mandarins(Citrus Reticulata Blanco) varieties.With 31 SRAP primers,which were polymorphic and uniformly distributed in mandarins genome,a total of 175 alleles were detected among the analyzed mandarins cultivars with the average number of alleles per pair of primers of 5.6452.Primers of Me 10-Em 11 and Me5-Em 11 generated the highest number of alleles,which were 11,and the Me8-Emll had the fewest alleles,which were 2.The genetic similarity among the mandarins cultivars was ranged from 0.58~0.94.Based on the SRAP markers, all the cultivars were divided into nine groups with the threshold of 0.785 of similarity coefficient.:1) Guangpi Da Guo Daoxian wild mandarin and Choupi Gan mandarin;2) Huapi Ju mandarin,Cazi Gan,Jiang Jin Sour orange,Indian sour orange,Shatangju mandarin,Ponkan mandarin,willow leaf mandarin,An Hua Hong Zao Su Mi Gan"mandarin,Changsha mandarin,Nanfenmiju;Miyamoto satsuma;3) Mangshan wild mandarin, Bendi Zao mandarin,Changsha mandarin,Xi Pi Gou Shi Gan,Changsha mandarin,Fuju,C.ichanggensis Swing;4) Limon;5) citron;6) Huapi Gan mandarin;7) ShaTian pummelo;8) Kumquat;9) Indian wild orange.Study of 32 pumelo and SRAP related species for analysis,clustering results show that:similar to coefficient of 0.68 for the critical value,can be divided into 32 samples of four groups,Shatian series,as the center and contains wendan pummelo Relatives lime series,sweet orange and wild-type resources.SRAP markers to distinguish between the various basic varieties(lines),all types of bands has its own characteristics.The relationships among the native citrus resources and pumelo germplasm in Hunan were established.
