巨峰葡萄查尔酮异构酶(CHI)基因的克隆及表达特性的半定量RT-PCR分析

【摘要】 试验从一年生巨峰扦插苗中克隆了CHI基因,并采用半定量RT-PCR技术,以actin基因为内参,分析了CHI基因在巨峰葡萄果实不同发育阶段及组织的表达。对CHI基因的序列分析表明:CHI基因序列中含有3内含子,4个外显子;生物信息学分析表明,CHI基因编码的多肽链既没有信号肽,也不存在跨膜结构域,且亚细胞定位于细胞质中。半定量RT-PCR分析结果表明,CHI基因在果皮、果肉、种子及幼叶和幼根中都表达,但不同的组织中表达方式不同。CHI基因在花后30和90天的果皮中强烈表达;但在果肉中,CHI基因在花后90天表达强烈,而在种子中于花后45天表达强烈,随果实发育,其表达逐渐下降。在上述实验的同时,高温处理抑制CHI基因在一年生盆栽巨峰葡萄幼叶中的表达,但随处理时间延长,其表达又有所恢复;高温诱导CHI基因在幼根的表达。

【Abstract】 CHI was cloned from one-year old cutting seedlings and sequence analysis showed that CHIcontained 3 intron and 4 extrons.Bioinformatics analysis indicated that a Signa peptide and transmembrane regions did not exsited in the multi-peptide encoded by CHI,and CHI subcellular localization was in cytoplasm.Expression analysis of CHI of Kyoho grape was carried out by semi-quantity RT-PCR, using grape actin gene as an internal control.The results showed that CHI expressed in berry skin,berry flesh and berry seeds,young leaves and roots,but had different expression patterns in different developmental stages and tissues.CHI expressed strongly at 30 and 90 days after flowering in berry skin;In berry flesh,CHI expressed strongly at 90 days after flowering;And at 45 days after flowering,CHI expressed intensively in berry seeds of Kyoho grape,then CHI expressed decreased rapidly with the berries developing.CHI expression was inhibited in young leaves and recovered with the treatment time longing,and induced in young roots in one- year-old potted cutting seedlings of Kyoho grape under high temperature.
