桃遗传多样性及亲缘关系的ISSR和SSR分析

【摘要】 利用ISSR和SSR标记对23份桃种质资源进行遗传多样性及亲缘关系检测。结果发现:18条ISSR引物共扩增出137条DNA片段,其中56条具有多态性,多态性频率为40.88%;每个ISSR引物可扩增出5~12条DNA片段,平均7.61条;材料间遗传相似系数GS变幅为0.60~0.91,平均为0.77。在供试材料具有多态性的8对SSR标记扩增出23条DNA片段,其中22条具有多态性,多态性频率高达95.65%;材料间遗传相似系数GS变幅为0.26~1.00,平均为0.62;ISSR标记能将其中21份桃种质完全区分开,SSR标记能区分开其中的16份。这两种标记的分子聚类结果大体吻合,都反映了多数来源地相同的种质表现出较为密切的亲缘关系。该研究结果对桃种质资源的保存和利用以及核心种质构建具有重要意义。

【Abstract】 The genetic diversity and genetic relationships among 23 peach accessions were evaluated by ISSR and SSR markers.In ISSR analysis,a total of 137 bands were detected,among which 56 bands were polymorphic.The number of bands from each ISSR marker ranged from 5 to 12,with an average of 7.88.In SSR analysis,a total of 23 bands was detected,among which 22 bands were polymorphic.The percentage of polymorphic bands detected by ISSR and SSR was 40.88%and 95.56%,respectively.The genetic similarity revealed by ISSR ranged from 0.60 to 0.91,with a mean of 0.77;and revealed by SSR ranged from 0.26~1.00, with a mean of 0.62.The genetic relationship of peach accessions were analyzed and 23 germplasm were classified on molecular level by clustered analysis,which indicated that ISSR could distinguish more peach cultivars than that SSR did.The grouping associations could be related to the mode of introduction and maintenance of the peach germplasm involving limited foundation germplasm.The results obtained in this work will help to improve the conservation and management of local China peach germplasm allowing the establishment of core collections that represent most of the diversity conserved.
