西瓜EST资源的SSR信息分析及标记开发

【摘要】 <正>本研究对5036条西瓜无冗余EST序列进行了搜索,共检索出350个SSR,检出率为6.09%,平均分布距离为7.43 kb。包括97种重复基元,其中二核苷酸和三核苷酸重复基元占主导地位,分别占总SSR的39.43%和46.86%。AT是二核苷酸中的优势基元,占二核苷酸重复类型的23.19%。三核

【Abstract】 In this study,5036 non-redundant ESTs of watermelon were screened and 350 SSRs were mined out.The frequency of these EST-SSRs was 6.09%and mean distance was 7.43 kb in the non-redundant ESTs.All these SSRs included 97 kinds of repeat motifs and dinucleotide and trinucleotide repeats were dominant types,accounting for 39.43%and 46.86%,respectively.AT is most frequent motif in Dinucleotide accounting for 23.19%.But the distribution of trinucleotide repeats was dispersive and the dominant motif was GAA only accounting for 8.54%in all the trinucleotide repeats.A total of 11 primer pairs were designed and tested against genomic DNA of watermelon.Eight primers could produce valid bands and the polymorphic bands were ranged from 2 to 6.All the eight primers were polymorphic between cultivated watermelon and wild watermelon germplasm.The above results indicated that watermelon EST-SSR was effective and feasible.
