肥胖与正常体重人群肠道菌群和UCP2基因多态性差异研究及其相互关系初探

【摘要】 目的研究成都地区单纯性肥胖和正常体重成人六种肠道菌群的数量差异及解偶联蛋白2基因(UCP2)ALa55Val多态性,初步探讨UCP2基因Ala55Val不同基因型与肠道菌群关系。方法采用培养法比较91例成都地区成人(单纯性肥胖组43例,正常体重组48例)六种肠道菌群的数量差异,通过PCR-RFLP检测研究对象UCP2基因ALa55Val多态性,并分别比较不同基因型间六种肠道菌群数量的差异。结果单纯性肥胖组与正常体重组A菌、B菌、C菌、D菌有显著差异(P<0.05)。两组均存在UCP2基因Ala/Ala,Ala/Val,Val/Val多态,其表型分布有显著差异(χ~2=12.21,P<0.05);单纯性肥胖组UCP2等位基因突变频率高于正常体重组,差异有统计学意义(χ~2=10.06,P<0.05);单纯性肥胖组、正常体重组和研究总体中不同基因型人群的六种肠道菌群数量无显著差异(P>0.05)。结论 A菌、B菌、C菌、D菌可能与肥胖有关。单纯性肥胖组与正常体重组的UCP2基因多态性及等位基因分布频率比较的差异表明,UCP2基因ALa55Val变异可能是成都地区单纯性肥胖的危险因素:对肠道菌群数量和种类与UCP2基因Ala55Val变异关系的探讨,提示该基因ALa55Val变异与肥胖的关联可能与肠道菌群数量改变无关。

【Abstract】 Objective To initially study on the viable counts of six gut bacteria,the polymorphism of UCP2 gene and its relationship with gut bacteria in Chengdu simple obesity and normal-weight people.Methods Culture methods were used to assess the viable counts of six gut bacteria of 91 Chengdu people(the simple obesity group,48 subjects; the normal-weight group,43 subjects).PCR-PFLP was applied to determine the genotypes of Ala55Val variant in the UCP2 gene of the two groups.And six kinds of gut bacteria among different genotypes in different groups were analyzed. Results The numbers of A,B,C and D of the simple obesity group were significantly lower as compared to the normal-weight group(P<0.05).Both the two groups had the Ala55Val variants of Ala/Ala,Val/Val and Ala/Val in the UCP2 gene,and the Ala55Val genotype distributions between the two groups was significantly different(x~2= 12.21,P<0.05).The allelic mutation frequency in the simple obesity group was higher than that of the normal-weight group(x~2 = 10.06,P<0.05).However,no significant difference was observed in the population of six gut bacteria among different genotypes in different groups(P>0.05).Conclusion A,B,C and D may be associated with obesity.The difference of Ala55Val variant in the UCP2 gene as well as the allelic frequency distributions between simple obesity group and the normal-weight group implied that the UCP2 gene mutation might be a risk factor of obesity in Chengdu area.The initial investigation on the relationship between the UCP2 gene Ala55Val variant and the gut bacteria indicated that this gene mutation may not be an impact factor on the alternation of gut bacteria.
