【摘要】 目的探讨老年男性肠脂肪酸结合蛋白(intestinalfatty acid binding protein,IFABP)基因exonⅡ54位点编码丙氨酸或苏氨酸(A/T)单核苷酸多态性(single nucleotidepolymorphism s,SNP)与血清血脂水平的关系。方法研究对象为114例老年男性,平均年龄71.3±7.2岁:对照组102例年轻男性,平均年龄38.2±3.1岁。利用聚合酶链反应(PCR),Hha I内切酶酶切,基因测序等技术检测各组IFABP基因型;血清总胆固醇(TC)、三酯酰甘油(TG)、高密度脂蛋白胆固醇(HDL-c)、低密度脂蛋白胆固醇(LDL-c)、载脂蛋白A I(Apo A I)、载脂蛋白B(Apo B)等指标的检测采用生物化学方法。结果老年男性组IFABP基因型A/A、A/T、T/T频率为0.29,0.53,0.18;对照组IFABP基因型A/A、A/T、T/T频率为0.52,0.35,0.13。突变型54T等位基因频率分别为:老年男性组0.45,对照组0.30。与对照组比较老年男性组54T等位基因频率明显增高,差异显著(x~2=9.41,p<0.01;)。在老年男性组,与54A等位基因携带者比较,54T等位基因携带者血清三酯酰甘油、低密度脂蛋白胆固醇、载脂蛋白B明显升高(分别为1.83±0 67 vs 1.48±0 67mmol/L,p<0.05:2 39±0.42 vs 2.21±0.41mmol/L.p<0.05:0.69±0.17 vs0.61±0.19g/L,p<0.05)。结论突变型54T等位基因频率在老年男性人群中明显升高,并且与老年男性较高水平的血清三酯酰甘油、低密度脂蛋白胆固醇、载脂蛋白B有关。更多还原

【Abstract】 Objective To investigate the association of frequency atcodon 54Ala/Thr(A/T) single nucleotide polymorphism (SNP) in the intestinal faty acid binding protein (IFABP) with serum lipids in agedmen.Methods Subjects were 114 elderly men aged 71.3±7.2 years Controls were 102 younger men aged 38.2±3.1 years.The Ala 54 Thrallelic variantwas detected by polymerase chain reaction, HhaIdigestion,and sequencing.TC,TG,HDL-c, LDL-c,ApoA I and ApoB in fasting serum were detected by biochemical technique.Results The genotype frequencyies of A/A,A/T,T/T were 0.29,0.53,0.18 in elderly men,0.52,0.35,0.13 in controls.The allelic frequency of pointmutant 54Thrin IFABP gene were 0.45 in elderly men,0.30 in controls,respectively Compared with controls, the allelic frequency of 54T in elderly men was markedly difference(x~2=9.41,p <0.01).The TG,LDL-c and ApoB concentraton in fasting serum in elderlymen with 54T allele were significantly higher than these with 54A allele (1.83±0.67 vs 1.48±0.67mmol/L,p <0.05;2.39±0.42 vs 2.21±0.41mmol/L,p <0.05;0.69±0.17 vs 0.61±0.19g/L,p<0.05).Conclusions In elderly men, the frequency ofmutantallele 54T was significantly elevented, whichwas associated with high TG,LDL-c and ApoB concentraton in fasting serum.更多还原
