应用离线二维液相色谱(分子筛×反相)分析发花前和发花后的茯砖茶样品

【摘要】 茯砖茶中的次生代谢成分极其丰富和复杂。由于其中含有很多潜在的生物活性化合物,所以通过一套系统的色谱方法尽可能多的分离分析这些成分就显得越来越重要。但是随着相关研究的不断深入,我们发现单一的高效液相法已不足以满足对其成分的检测需要。二维液相色谱因其高分辨率及高峰容量逐渐应用于复杂基质样品的分析检测中。本文采用离线二维液相色谱(2-D LC)的方法进行茯砖茶发花前后的成分分离检测,其中第一维采用排阻色谱柱(SEC),第二维采用反相色谱柱(RP),依次根据化合物的分子量及极性进行分离。2-D LC因两根柱子之间的低相关性(r2<0.2)而表现出较大的峰容量(>1800)。该方法明显的改善了那些在传统HPLC上无法检测到的以及共洗脱的化合物的分离分析效果。
【Abstract】 The complex secondary metabolites in Fuzhuan brick-tea are potential for bioactivity discovery. It is important to analysis these compounds as complete as possible in one chromatographic system, which beyond the capacity of single liquid chromatography(LC). On the other hand, with both high resolution and high peak capacity, more two dimensional LC(2-D LC) methods are being applied to analysis the samples of complex chemical matrix. In this report, an off-line LC × LC system was designed to separate the before and after fungal fermented Fuzhuan brick-teas(AFFT and BFFT), based on size exclusion chromatography(SEC) in the first dimension, and reversed phase(RP) in the second dimension. The designed 2-D LC system was characterized by a very high practical peak capacity(> 1800) due to the low degree of correlation(r2 < 0.2) between the selected separation mechanisms. Furthermore, this method demonstrated its suitability for analysis of Fuzhuan brick-tea, by clear exhibition of the peaks of some interesting compounds, which are co-eluted in single dimensional LC of either RP or SE mechanism.
