近岸海水中十溴联苯醚抗性菌的筛选、分离及鉴定

【摘要】 从受十溴联苯醚污染的海水中富集筛选得到十溴联苯醚的抗性菌株,旨在为受污染的土壤和海水的生物修复提供菌种资源。选用受十溴联苯醚污染严重的海水为种源,以十溴联苯醚为唯一碳源,经过多次富集、分离、纯化至单菌落,观察各个菌落的形态、颜色,再进行细菌基因组DNA的提取、PCR扩增、产物纯化,纯化后的产物进行16SrDNA测序。测序结果经Eztaxon比对,结合菌落的形态判断其种属。结果显示:以十溴联苯醚为唯一碳源,从泉州和东山两个海水样品中共筛出两组13株十溴联苯醚的抗性菌株。这13株细菌分属于10个属,包括芽孢杆菌属、弧菌属、假单胞菌属等。其中D-5与模式菌株Aequorivita capsosiphonis A71(T)的相似度仅为97.500%,D-7与模式菌株Novosphingobium pentaromativorans US6-1(T)的相似度仅为97.609%,这两株菌可能为新种。结论实验所得13株十溴联苯醚抗性菌,可能具有十溴联苯醚的降解功能,D-5和D-7可能编码了一种新的降解基因,其降解效率有待进一步研究。
【Abstract】 Decabromodiphenyl ether resistant bacteria was enriched and screened from polluted water of decadromodiphenyl ether, which was used to bioremediation of polluted water and soil. Decabromodiphenyl ether resistant bacteria was isolated from polluted water by selective enrichment culture in the experiment condition. The strains were able to grow on decabromodiphenyl ether as a sole carbon source. They were identified based on their morphological and 16S rDNA sequence analysis. Results The thirteen bacteria, which were able to use decabromodiphenyl ether as the sole carbon source, were isolated from seawater of Quanzhou and Dongshan. They belonged to ten species,including Bacillus, Vibrio, Pseudomonas etc. The similarity of D-5 and Aequorivita capsosiphonis A71(T) was only 97.500%. The similarity of D-7 and Novosphingobium pentaromativorans US6-1(T) was only 97.500%.The two strains may be new species.The thirteen bacteria may be degradation of decabromodiphenyl ether function. D-5 and D-7 are likely to encode a new degradation gene. More research should be performed on the degradation efficiency to confirm whether they are be used to degrade the bioremediation of marine electronic waste pollution.
