日本鳗鲡不同组织或器官的抗菌活性比较
Antibacterial Activity in Different Tissues or Organs of Japanese Eel,Anguilla Japonica

【摘要】 用10%醋酸提取日本鳗鲡肝脏、鳃、脾脏、胆汁、肾脏和血清中的抗菌活性物质,检测其对养殖鳗鲡常见的四种致病菌,即肠杆菌Enterobacter sp.B01、迟缓爱德华氏菌Edwardsiella tarda B09、气单胞菌属Aeromona sp.B18和嗜水气单胞菌Aeromona hydrophila B27)抑杀作用。结果显示,肝脏、脾脏、胆汁和鳃的样品对四株试验菌株均有明显的抑菌作用,肾脏对B18、B27和B01三株致病菌有抑菌作用,血清仅对B18有抑菌作用,不同样品对同一种致病菌的抗菌活性差异显著(p<0.05),按平均抑菌圈半径计算,其抗菌活性大体呈现为:肝脏>鳃>脾脏>胆汁>肾脏>血清。同一样品对不同细菌的抑杀作用差异显著(p<0.05)。采用反相高效液相色谱法分析上述各组织/器官的醋酸提取物中的活性组分,结果显示:肝脏、脾脏、胆汁和鳃样品中的活性组分(以杀伤指数大于50%计)主要集中于乙腈洗脱浓度为19.4%-35.4%,肾脏和血清中未分离出抗菌活性组分(杀伤指数小于50%)。本研究为以后研究鳗鲡等鱼类抗菌活性组分提供重要的参考依据。
【Abstract】 A search for antibacterial activity in different tissues/organs including liver, gill, spleen, bile, kidney and serum of Japanese eels (Anguilla japonica) was conducted. In this study, we obtained extract crudes of those tissues/organs from Japanese eels. Antibacterial activity against four common pathogenic bacteria from eels including Enterobacter spp B01, Edwardsiella tarda B09, Aeromonas spp B18 and Aeromona hydrophila B27 was detected by using agar diffusion method. The results showed that antibacterial activities of liver, gill, spleen, bile are obvious. The crudes of kidney against B18, B27, B01 and serum against B18 are effective. Antibacterial activity varied of different tissues/organs against the same bacterium. The antibacterial activity of liver is highest. The antibacterial activity varied of the same tissues/organs against different bacteria. The acid extractions were loaded onto reverse-phase liquid chromatography on Source 5R RPC-ST-4.6/150 column(Amersham Bioscience, Sweden). The results showed that liver, kidney, gill, bile the killing indexes of which higher than 50% are distribute from 19.4% to 35.4% .The killing index of serum and kidney eluates are lower than 50%.
