紫茎泽兰草粉致突变致畸作用研究

【摘要】 目的:探讨紫茎泽兰草粉的致突变致畸作用。方法:紫茎泽兰草粉小鼠骨髓嗜多染红细胞微核试验及小鼠睾丸精母细胞染色体畸变试验均以10.0、5.0、2.0g/kg剂量经口给予5天,观察骨髓嗜多染红细胞微核率及睾丸精母细胞染色体畸变率。Ames试验选用TA97、TA98、TA100和TA102四个菌株,试验剂量为5000、2500、1250、625、312μg/皿,观察各菌株的回变菌落数。大鼠致畸试验按照《饲料安全性评价喂养致畸试验》方法,将紫茎泽兰草粉参入饲料中,剂量为10.0、6.0、3.0g/kg。观察孕鼠的生殖情况和胎鼠的生长发育、骨骼内脏畸形情况。结果小鼠骨髓嗜多染红细胞微核率及小鼠睾丸精母细胞染色体畸变率与阴性对照组比较,均无显著性差异(P>0.05)。Ames试验,紫茎泽兰草粉各剂量组的回变菌落数均未超过自发回变菌落数的两倍,亦未见剂量-反应关系,Ames试验结果为阴性。致畸试验各剂量组孕鼠进食、活动、体重增长、死胎率、吸收胎率均未见明显影响;胎鼠的体重、身长、尾长与阴性对照组相比无明显差异(P>0.05);胎鼠外观、骨骼、内脏检查均未发现畸形。结论紫茎泽兰草粉无致突变致畸作用。

【Abstract】 Objective:To study the mutagenicity and teratogenicity of Eupatorium adenophorum powder. Methods The mice were gavaged Eupatorium adenophorum powder at dose of 10.0、5.0、 2.0g·kg-1·d-1 for 5 days. Chromosome aberration of spermatocytes and micronucleus of bone marrow were tested. Strains of TA97、TA98、TA100、TA102 were employed in Ames test. The exposure concentraton is 5000、2500、 1250、625、312μg/plate. According to feed safety evaluation feeding teratogenicity test, Eupatorium adenophorum powder was mixed with feed.The rats were gavaged Eupatorium adenophorum powder at dose of 10.0、6.0、3.0g·kg-1·d-1. Embryonic and fetal developmental parameters were surveyed. External, skeletal and internal organ anomalies were examined. Results No significant difference were found in chromosome aberration and micronucleus rate between control and experimental groups. Ames test(in presence or absence of S9) showed no two fold increase in the spontaneous reversion colonies.There was no dose-dependence in the number of revertant colonies either.The result of Ames test is negative. Compared with negative control group, No significant difference were found in eating,activity, weight gain in all treated groups. There were no difference in the rate of fetal mortality and embryos absorption in all treated and negative control group. The weight, body length and tail length showed no significant difference between the treated groups and negative control group. No external and internal organ and skeletal anomalies were observed.Conclusion The results suggested that Eupatorium adenophorum powder had no mutagenic and teratogenic effects.
