体外酶促氧化合成茶黄素研究

【摘要】 以政和大白茶鲜叶匀浆液为酶源,在比较研究政和大白茶、秀红、碧香早三种鲜叶中的儿茶素合成茶黄素的基础上,采用单因素试验、正交试验、验证试验的方法,系统优化筛选了20g最优品种鲜叶原料酶促合成茶黄素的酶源用量、酶促反应时间、温度、pH值等技术参数,探讨了在该技术参数下利用速溶绿茶、90%茶多酚作为底物合成茶黄素的效果,以为茶黄素的高效制备提供技术支持与数据参考。结果表明,政和大白茶鲜叶原料中的儿茶素最适合于合成茶黄素,20g鲜叶原料合成茶黄素3.78mg,同时,从酶促反应前后的儿茶素变化来看,参与茶黄素合成的儿茶素主要是EGCG、ECG、EGC、EC;20g政和大白茶鲜叶原料合成茶黄素的最优技术参数为酶源用量0.6g/100mL、酶促反应时间120min、温度35℃、pH值5.2,在该参数下,茶黄素的合成量达5.10mg以上;利用速溶绿茶可有效合成茶黄素,且24g速溶绿茶可合成17.23mg茶黄素。

【Abstract】 Based on fresh leaves from Zheng He Dabai as enzyme and materials from Zheng He Dabai,Xiu Hong and Bi Xiangzao to synthesize theaflavins was researched.the single factor,orthogonal experiments and verifying test were adopted to select with system optimization such technology parameter as the dosage of enzyme,the time of enzymatic reaction,temperature,pH value and so on.And the effect of using instant green tea and polyphenols whose purity is 90% as substrate to synthesize theaflavins was discussed under the technology parameter,which can provide technology support and data reference for preparing theaflavins efficiently.The results indicated that the catechins in fresh leaves of Zheng He Da Bai was the best to synthesize theaflavins.20 gram fresh leaves can synthesize 3.78 milligram theaflavins,and the main catechins of participating in synthesizing theaflavins were EGCG,EGC,EC and ECG.The optimum technology parameter of 20 gram fresh leaves of Zheng He Da Bai to synthesize theaflavins was that the dosage of enzyme was 0.6g/100mL,the time of enzymatic reaction was 120 minutes,temperature was 35 ℃,pH value was 5.0.Under the parameter the quantity of synthesizing theaflavins can reach more than 5.10mg.
