【摘要】 MicroRNAs(miRNAs)是基因组编码的小分子非编码RNAs,在生长发育、生物和非生物胁迫以及其他方面均起到重要作用。为解析MicroRNAs表达调控,本研究利用TSSP程序和PlantCARE数据库预测了来自大豆降解组文库的440 miRNA的核心启动子以及369 miRNAs的顺式作用元件。结果表明:440 miRNA中有83.86%在其上游序列中包含启动子,8.64%(38个位点)在下游序列中预测到1个启动子。核心启动子的TATA盒与转录起始位点(TSSs)的分布相似,在转录起始位点100 bp区域内分布较集中。此外,对转录起始位点5’的顺式作用元件预测为miRNAs的可能功能和表达的时空性提供了线索。对miRNAs的顺式作用元件和目标基因的分析显示在miRNA的表达与生长素响应因子、赤霉素响应因子之间存在潜在的负反馈调控回路。本研究为大豆miRNAs功能和表达调控研究提供了重要信息。

【Abstract】 MicroRNAs(miRNAs) are genome-encoded,small non-coding RNAs that play important functions in development,biotic and abiotic stress responses,and other processes In this study,to examine the regulation of miRNA gene expression,we used bioinformatic program TSSP and PlantCARE database predict the core promoters of 440 miRNAs identified from a soybean degradome library and found cis-acting elements for 369 miRNAs.The prediction results showed that 83.86%of the 440 miRNAs contained promoters in their upstream sequences,and 8.64%(38 loci ) were predicted to have a promoter in their downstream sequences.The distribution of two core promoter elements,TATA-boxes and transcriptional start sites(TSSs),were similar,and distributed intensively in the transcription start site 100 bp region.Moreover,the cis-acting elements 5’ of the TSSs were examined to obtain clues to potential functional and spatiotemporal expression of the miRNAs.Analysis of miRNA cis-elements and targets indicates a potential negative feedback loop involving miRNA expression and auxin response factors and CA response factors.The data obtained here provide insights into the transcription regulation and functions of miRNAs in soybean.
