高赖氨酸转基因玉米的鉴定和检测

【摘要】 将玉米22-KD醇溶蛋白基因干涉载体采用基因枪轰击的方法转化到玉米幼胚愈伤组织中,对其后代PCR检测发现,T0代最终获的可以结实的阳性转基因事件数为9个;T1代阳性株系的赖氨酸含量检测结果表明,转基因株系与对照相比变幅较大,在-12.2%-63.6%之间,其中2个株系的赖氨酸含量达到了优质蛋白玉米的标准(0.419和0.468),分别比对照提高了46.5%和63.6%;阳性籽粒的22-KDα-醇溶蛋白与非QPM材料B73、云瑞21和阴性对照相比下降明显,说明转导的22-KD干涉基因在一定程度上抑制了22-KD基因家族的表达。
【Abstract】 22 - KD zein gene interfering vectors of maize were transferred into maize immature embryo callus by gene gun bombardment in this study,and their offspring were tested,achieved the following results:9 positve events were harvested by PCR test in the T0 generation。The changing range of Lysine content in T1 generation were very wide from - 12.2% ~63.6% ,and 2 lines reached the lysine content of the standard of quality protein maize(0.419 and 0.468),which increased 46.5% and 63.6% respectively; the 22 - KD alfa - gliadin of positive transgenic grains were significantly reduced compared to non - QPM materials B73,YR21 and negative control,which indicated that the 22 - KD interfering gene suppressed the expression of the 22 - KD gene family.
