鱼、鸭样品中微囊藻毒素含量测定方法研究

【摘要】 目的建立鱼、鸭肉、肝脏样品中微囊藻毒素(MC-LR)含量的测定方法。方法取鱼、鸭肉、肝脏经组织捣碎机捣碎,甲醇提取,离心,Sep-pakC18(ODS)小柱富集、净化,0.1%三氟乙酸(TFA)甲醇梯度解析,酶联免疫吸附(ELISA)法测定含量。结果该方法预处理样品分离效果好,对测定无干扰,方法最小检出浓度为0.1ng/g,相对标准偏差为5.34%,加标回收率为90%~92%。结论所建立的方法经实际应用取得了满意的效果。

【Abstract】 Objective:Indirect competitive enzyme-linked immunosorbent assay(ic-ELISA) was established to detect microcystin-LR in fish and duck samples.Method:Hepatic tissue of the fish and ducks,meat of fish and duck were triturated by blender,extracted with methanol,centrifuged,enriched by Sep-pakC18(ODS),purified,gradient elution separated with 0.1% trifluoroacetic acid(TFA) methanol,detected microcystin-LR using Enzyme-linked immunosorbent assay(ELISA).Results:Minimum detectable concentration is 0.1ng/ g,relative standard deviation is 5.34%,recovery rate is 90%~92%。 Conclusion:This method performed well on pre-process for sample separation and it was under the influence from the samples.
