【摘要】 制备载Survivin siRNA的壳聚糖纳米粒,以期实现使喉癌癌细胞凋亡作用。用复凝聚法制备载SurvivinsiRNA的壳聚糖纳米粒,用琼脂糖凝胶电泳分析质粒的纯度和纳米粒的电性结合,TEM观察纳米粒的粒径和形态,计算该纳米粒的包封率。采用复凝聚制备法能够实现壳聚糖对的包埋,壳聚糖与Survivin siRNA电性结合稳定,包封率达到92.8%,制备的壳聚糖质粒纳米粒呈亚球型,粒径为50-100nm。

【Abstract】 To prepare chitosan nanoparticles loaded with Survivin siRNA and to accomplish the death of cancer cells by apoptosis.Survivin siRNA was loaded into chitosan nanoparticles by using the complex coacervation method. The purity quotient of the plasmid and the integration of the nanoparticles were analyzed by agarose gel electrophoresis,the grain diameter and shape of the nanoparticles was determined by TEM.The encapsulation ratio was also calculated.The chitosan nanoparticles loaded with Survivin siRNA were successfully prepared.Chitosan and Survivin siRNA was integrated stably by electrostatic interaction.The nanoparticles with encapsulation ratio of 92.8%were prepared.The nanoparticles were nearly spherical in shape with the diameter of 50-100 nm.
