不同施肥条件下毛竹根系内生细菌的分离及其多样性分析

【摘要】 植物内生菌广泛分布于植物体内,其多样性受宿主植物类型和外部环境因素的影响。采用平板培养方法对三种不同施肥状态的毛竹根系内生菌进行分离计数,随机挑取其中150个菌落,依形态特征进行初步归为11类,进而以细菌的通用引物扩增16S rDNA片段,用两种限制性内切酶HhaⅠ和RasⅠ酶切,对限制性酶切多样性进行PCR-RFLP分型后测序。测序结果表明,毛竹根系内生菌归于芽孢杆菌属(Bacillus),肠杆菌属(Enterobacter),假单胞菌属(Pseudomonas),贪铜菌属(Cupriavidus),微球菌属(Micrococcus),金黄杆菌属(Chryseobacterium)六个属。施肥状况不同,内生细菌的种类和数量也不同,占主要优势的群落类型也发生改变。
【Abstract】 Endophytic bacteria widely existed in plants and the diversity was effected by the host plants and environment.In this paper,150 endophytic bacteria were obtained separately from the roots of Phyllostachys edulis by plate culture method.The 150 endophytic bacteria were categorized preliminarily to 7 groups based on colony morphology.16S rDNAs were amplified with universal primers of bacteria,the PCR products were digested with restriction endoenzyme HhaⅠand RsaⅠrespectively and analyzed by RFLP analysis.The results showed that Endophytic bacteria content was 10~6 cfu/g.There were 7 restriction types and the main type accounted 48%. 16S rDNA sequencing indicated that Pseudomonas sp.was the major genera.
