海带多糖对2型糖尿病小鼠血清胰岛素和胰淀素水平的影响

【摘要】 目的探讨海带多糖在实验性2型糖尿病小鼠中的降血糖作用和可能的机制。方法健康雄性昆明小鼠60只,高脂饲料喂养并腹腔注射四氧嘧啶制备2型糖尿病动物模型,昆布多糖灌胃干预治疗。自动血糖仪测小鼠空腹血糖(FBG),免疫电化学发光法检测血清胰岛素(Insulin)水平,酶联免疫吸附法检血清胰淀素(Amylin)水平。结果经海带多糖干预治疗后,高-中-低剂量组动物FBG水平较模型组显著降低(P<0.05);血清胰岛素水平和Amylin水平较模型组显著升高(P<0.05);各剂量组之间无明显差异(P>0.05)。结论海带多糖在2型糖尿病早期能促进胰岛细胞的分泌功能,提高血清胰岛素和胰淀素水平,发挥降糖作用。
【Abstract】 Objective To investigate the hypoglycemic effect and possible mechanism of Laminaria Japonica polysaccharide(LJPS)in type 2 diabetes mellitus model mice.Methods Type 2 diabetes mellitus models were established by feeding high fatty forage and injecting alloxan in 60 healthy male mice.The LJPS was applied as additive in physiological saline to treat the mice by irrigation.The levels of fasting blood glucose(FBG)were detected by automatic blood glucose device.The electrochemical immunological assay was applied to determine the insulin level in serum.The enzyme-linked immunosorbent assay(ELISA)was applied to determine the amylin level in serum.Results After treated with LJPS,the FBG in the low-middle-high dosage groups decteased significantly than those in the model group(P<0.05);while the levels of insulin and amylin in serum increased significantly than those in the model group(P<0.05).There were no significant differences among the low,middle and high dosage groups all above indexes(P>0.05).Conclusion LJPS have hypoglycemic effects in early period of type 2 diabetes mellitus by promoting the islet cell to secret insulin and amylin.
