利用MAR元件提高禽流感血凝素基因在百脉根中表达的研究

【摘要】 【目的】利用豆科牧草作为植物生物反应器来生产禽流感抗原蛋白,研究提高抗原蛋白表达量的方法。【方法】构建含高效表达元件MAR序列的植物高效表达载体,以百脉根的子叶作为外植体,通过农杆菌介导法,将禽流感血凝素基因导入到了豆科牧草推广品种百脉根"里奥"中,经过抗性筛选,获得了抗性植株,对抗性植株进行RT-PCR、Southern Blot、Western Blot、Elisa检测分析。【结果】检测结果表明禽流感血凝素基因已成功在百脉根中表达,MAR元件可以提高禽流感血凝素基因在百脉根中的表达。【结论】利用豆科牧草作为植物生物反应器来生产禽流感抗原蛋白是切实可行的。

【Abstract】 [Objective]Legume forage was used as a plant bioreactor to produce the H5N1 antigen protein.In the study,we explored the method for improving the expression level of H5N1 antigen protein. [Method]One high effective plant expression vectors with MAR sequence was constructed by genetic recombination technology,and transformed into Lotus corniculatus.The hemagglutinin gene of avian flu virus was tranfered into the legume plant Lotus corniculatus"Leo"by Agrobacterium-mediated transformation,After selective differentiation and regeneration,positive plants were obtained.Positive plants were detected by RT-PCR,Southern blotting and western blotting.[Result]The results has proved that the avian infleuenza hemagglutinin gene was successfully expressed in Lotus corniculatus.. [Conclusion]It is feasible to use plant bioreactor to produce H5N1 antigen protein in the legume forage.
