多花黑麦草斑驳病毒外壳蛋白基因在大肠杆菌中的表达

【摘要】 根据已报道的多花黑麦草斑驳病毒(RMotV)外壳蛋白(CP)基因的核苷酸序列合成引物,用RT-PCR的方法获得外壳蛋白基因后,再将其克隆至pGEX-4T-1中,SDS-PAGE和斑点杂交(Dot-blotting)表明:经1PTG诱导,CP基因在火肠杆菌BL21(DE3)plys中获得了高效正确的表达,并且表达产物以不溶性包涵体的形式存在。
【Abstract】 Based on published nucleotide sequence,coat protein gene of RMotV(Ryegrass Mottle Virus,RMotV) was synthesized by reverse transcription polymerase chain reaction (RT-PCR) and constructed into expression vector pGEX-4T-1.The result of SDS-PAGE showed that successful expression of a 50kD fusion protein was achieved by IPTG induction,and furthermore it was indicated by Dot-blotting that the fusion protein was expressed in the form of insoluble inclusion bodies
