苯接触工人尿中苯巯基尿酸检测方法研究

【摘要】 目的探讨苯接触工人尿中苯巯基尿酸(S-PMA)的高效液相色谱测定方法。方法采用C18柱进行分离,以乙腈作为流动相A,乙腈-甲醇-三乙胺为流动相B进行梯度洗脱,流速2ml/min,柱温35℃,紫外检测波长205nm,尿样酸化后用5ml氯仿:异丙醇(5:1,v/v)萃取两次,氮气流挥干,残渣用100ul流动相B相溶解后进样分析。结果所建立的标准曲线在S-PMA的质量浓度为62.5~2000μg/L时线性关系良好(r=0.9994),加标回收率为100.56%~115.23%,日内与日间测定的相对标准偏差分别为3.4%~4.9%和2.3%~4.2%。结论该方法简便、灵敏,可用于职业苯接触者的生物监测。
【Abstract】 Objective To establish a method for determination of urinary S-phenylmercapturic acid in worker exposed to benzene by high performance liquid chromatography.Methods Separation was carried out on a C18 column by gradient elution with acetonitrile(A) and acetonitrile-methanol-triethylamine(B) as mobile phases, flowing at 2.0ml/min,UV detection at 205 nm and column temperature maintained at 35℃.Urinary sample was acidified with sulfuric acid and extracted twice using 5ml chloroform-isopropanol(5:1,v/v).The solvent was evaporated under a stream of nitrogen,the residue was redissolved in 100μl of mobile phase B for HPLC injection. Results Good linearity was observed within the range from 62.5μg/L to 2000μg/L(r=0.9994) and the detection limit was 40μg/L.The average recoveries for S-PMA were 100.56%-115.23%.Relative standard deviations(RSD) for intra-day and inter-day determinations were 3.4%-4.9%and 2.3%-4.2%,respectively.Conclusion This analytical method for S-PMA is simple,sensitive and convenient.It is suitable for the biological monitoring of occupational exposure to benzene.
