DGGE技术分析接种菌剂在猪粪堆肥中的动态变化

【摘要】 通过PCR-DGGE技术研究接种菌剂和自然发酵的猪粪堆肥中细菌群落的动态变化。结果表明,自然发酵的猪粪堆肥中土著细菌的种类较丰富,到堆肥结束时,与接种菌剂的堆肥相比其细菌群落组成变化不大;而在堆肥中接种菌剂后,堆肥第2天就成为高温期中的优势菌群,提高了堆肥的温度,延长了高温持续期,并且使原有的微生物群落结构发生较大的改变。研究表明堆肥中添加菌剂是提高堆肥温度和加快堆肥腐熟的有效方法,应用PCR-DGGE技术可以很直观准确地研究添加菌剂在堆肥中的动态变化。
【Abstract】 Microbial dynamics of microbial community during inoculated and natural composts were investigated with PCR technology and the methods of denaturing gradient gel electrophoresis(DGGE).The results indicated that natural composts had a variety of microorganisms;the microbial community,compared with inoculated compost changed little until the test completed.However,the inoculated strains assumed predominance during the high temperature phase after inoculating,and the microbial community had changed a lot.The result shows that inoculating is an effective way to raise temperature and shorten the time of composting;PCR-DGGE can help study Microbial dynamics of inoculants exactly.
