Matrigel基质胶对移植后的乳牙牙髓作用的动物模型的研究

【摘要】 目的:在裸鼠皮下移植于不同长度的牙根来研究牙根长度对裸鼠存活的影响,以及探求Matrigel基质胶对体外移植的乳牙牙髓存活率的作用。为后期乳牙牙髓自体移植至牙髓坏死的年轻恒牙根管中的实验提供相应的实验基础。方法一:收集2019年4-6月在南昌大学附属口腔医院因为正畸治疗而选择拔除的健康下颌前磨牙60颗,将实验分成5组,分别制备成6mm、7mm、8mm、9mm、10mm不同长度的牙根,根尖孔预备至2mm,冠部用MTA严密封闭。选取6周龄雌性裸鼠30只,植入已预备的牙根,6周后取出从裸鼠皮下取出牙根,进行组织学评估。方法二:收集2019年7-9月在南昌大学附属口腔医院因乳牙滞留而拔除的乳牙,选取牙根长度吸收不超过1/2的乳牙,获取相对完整的乳牙牙髓,乳牙牙髓低温保存于营养液中,快速移植于预备好的牙空管中,而后植入裸鼠皮下。实验分为3组,每组选取12只裸鼠,每组12颗乳牙。A组:乳牙牙髓+Matrigel+半牙空管;B组:乳牙牙髓+半牙空管组;C组:牙空管;4周后取出半牙空管,观察根管内容物并做组织学评价。结果:实验一通过HE染色显示不同长度的牙空管内长入新生组织以及各组裸鼠的愈合无明显差别。实验二:通过HE染色以及CD31染色表明实验组Matrigel+乳牙牙髓的移植后的乳牙牙髓处于存活状态,并且乳牙牙髓保持活性的数量明显高于其他对比组.结论:不同牙根长度在裸鼠皮下对于裸鼠伤口愈合无明显影响;Matrigel基质胶对于乳牙牙髓的体外移植的存活率具有明显促进作用

【Abstract】 Objective : To study the effect of root length on immunodeficiency mice by transplanting root canals with different root lengths in immunodeficiency mice subcutaneously,and to explore the effect of Matrigel on dental pulp in vitro.It will provide a corresponding experimental basis for the experiment of autogenous transplantation of the latex teeth into the root canal of young permanent teeth with pulp necrosis.Method 1 : sixty healthy mandibular premolars extracted from orthodontic treatment at the Affiliated Stomatological Hospital of Nanchang University from April to June 2019 were collected.The experiments were divided into 5 groups and prepared into 6mm,7mm,8mm,9mm,and 10 mm different lengths The root and apical foramen were prepared to 2mm,and the crown was tightly sealed with MTA.Thirty female 6-week-old immunodeficiency mice were selected,and the prepared roots were implanted.After 6 weeks,the roots were removed for histological evaluation.Method 2: Collect deciduous teeth extracted from deciduous teeth at the Affiliated Dental Hospital of Nanchang University from July to September 2019.Select deciduous teeth whose root length is not more than 1/2.The nutrient solution was quickly transplanted into an empty tooth tube of appropriate length.The experiment was divided into 3 groups,with 12 immunodeficiency mice in each group.Group A: deciduous tooth pulp + Matrigel + dental empty tube;group B: deciduous tooth pulp + dental empty tube group;group C: dental empty tube;after 64 weeks,remove the empty dental tube,observe the root canal content and do histological evaluation.Results : Experiment 1 showed by HE staining that there was no significant difference in the growth of new tissues in the dental canals of different lengths and the healing of nude mice in each group.Experiment 2: HE staining and CD31 staining showed that the dental pulp of the experimental group after Matrigel + deciduous dental pulp transplantation was alive,and the number of deciduous dental pulp retention activity was significantly higher than that of other control groups.Conclusion: Different root lengths under the skin of nude mice have no obvious effect on wound healing in nude mice.Matrigel significantly promotes the survival rate of deciduous dental pulp in vitro
