【摘要】 目的:本研究通过qRT-PCR技术检测胃间质瘤患者及健康人血浆外泌体中miRNA-423-5p的表达情况,并分析其与临床资料之间的关系。方法:选取2020年10月~2022年02月安徽医科大学第二附属医院确诊的胃间质瘤患者48例,其中女性18例,男性30例,对照组健康人10例。采用超速过滤方法提取胃间质瘤患者和健康人血浆中外泌体。通过透射电子显微镜、NTA、Western Blot对提取的血浆外泌体特性进行鉴定。通过q RT-PCR检测血浆外泌体中miRNA-423-5p的表达水平。收集胃间质瘤患者的相关临床资料,其中包括患者性别、年龄、肿瘤大小、核分裂象和危险度分级。并使用SPSS26.0统计学软件分析表达水平和临床资料之间的关系,计量资料使用(x±s)表示,组间比较用t检验,P(27)0.05为差异有统计学意义。结果:胃间质瘤患者血浆外泌体中miRNA-423-5p的表达水平较正常对照组明显升高(P<0.05),其表达水平与患者性别、年龄无关(P>0.05),与肿瘤直径、核分裂象、危险度呈正相关(P<0.05)。结论:miRNA-423-5p在胃间质瘤患者外泌体中表达明显上调,差异有统计学意义。与肿瘤直径、核分裂象及危险度呈正相关,差异有统计学意义。并且高表达的miRNA-423-5p和间质瘤预后不佳因素相关,提示外泌体miRNA-423-5p可能与预后不佳相关,并可能成为胃间质瘤诊断和判断预后的标志物。更多还原

【Abstract】 Objective: In this study,the expression of miRNA-423-5p in plasma exosomes from Gastric stromal tumor patients and healthy humans was detected by q RT-PCR,and the relationship between it and clinical data was analyzed.Methods: A total of 48 patients with gastric stromal tumor diagnosed in the Second Affiliated Hospital of Anhui Medical University from October 2020 to February 2020,including 18 female,30 male and 10 healthy patients in the control group.Plasma exosomes were extracted from gastric stromal tumor patients and healthy people by the overspeed filtration method.The characteristics of the extracted plasma exosomes were identified by transmission electron microscopy,Nanoparticle Tracking Analysis,and Western Blot.The expression levels of miRNA-423-5p in plasma exosomes were determined by real-time quantitative PCR.Relevant clinical data were collected from patients with gastric stromal tumor,including patient sex,age,tumor size,nuclear division image,and risk grade.The relationship between expression level and clinical data was analyzed using SPSS26.0 statistical software,measured using data(x ±s),and the group was compared by t-test.P(27)0.05 was considered to be statistically significant differences.Results: The expression levels of miRNA-423-5p in plasma exosomes were significantly increased in gastric stromal tumor patients compared with normal controls(P<0.05),and its expression level was not correlated with patient sex and age(P> 0.05),and positively correlated with tumor diameter,mitotic image and risk(P <0.05).Conclusion: The expression of miRNA-423-5p was significantly up-regulated in exosomes of gastric GISTs.There was a positive correlation with tumor diameter,nuclear division image and risk.Moreover,high expression of miRNA-423-5p and poor prognosis factors are associated with stromal tumors,suggesting that exosomal miRNA-423-5p may be associated with poor prognosis and may become a marker for the diagnosis and prognosis of gastric stromal tumor.更多还原
