HDAC2和乙酰化p53k320在结肠癌中的表达及与临床病理相关性研究

【摘要】 目的:检测HDAC2和乙酰化p53k320在结肠癌中的表达量,分析其与临床病理参数之间的关系及两者在结肠癌中的相关性,为结肠癌的治疗提供新的思路。方法:根据排除标准选择结肠癌患者收集癌组织标本,征集正常志愿者收集正常结肠黏膜组织,采用Western-blot法检测结肠癌组织及正常结肠黏膜组织中HDAC2和乙酰化p53k320的表达量;采用卡方检验对结肠癌组织中HDAC2和乙酰化p53k320在不同临床病理参数的表达差异进行比较;采用Person相关性分析对结肠癌组织中HDAC2和乙酰化p53k320的相关性进行分析。结果:(1)结肠癌组织中HDAC2的表达量为1.6±0.04,正常结肠黏膜组织中HDAC2的表达量为0.29±0.06,与正常结肠黏膜相比,结肠癌组织中HDAC2的表达上调,差异具有统计学意义(P<0.05);(2)结肠癌组织中乙酰化p53k320的表达量为0.1±0.01,正常结肠黏膜组织中乙酰化p53k320的表达量为0.78±0.02,与正常结肠黏膜相比,结肠癌组织中乙酰化p53k320的表达下调,差异具有统计学意义(P<0.05);(3)HDAC2和乙酰化p53k320在结肠癌中的表达与肿瘤大小(P<0.05)、结肠癌的分化程度(P<0.05)、有无淋巴转移(P<0.05)、TNM分期(P<0.05)有关;(4)HDAC2和乙酰化p53k320在结肠癌组织中的表达呈负相关(P<0.05、r=-0.364)。结论:(1)与正常结肠黏膜组织相比,HDAC2在结肠癌组织中呈高表达,乙酰化p53k320在结肠癌组织中呈低表达;(2)肿瘤越大、分化程度越低、伴有淋巴转移及TNM分期越晚,HDAC2的表达越高,乙酰化p53k320的表达越低;(3)HDAC2与乙酰化p53k320在结肠癌组织中的表达呈负相关(P<0.05、r=-0.364)。

【Abstract】 Objective:To detect the expression of HDAC2 and acetylated p53k320 in colon cancer,analyse their relationship with clinicopathological parameters and their correlation in colon cancer,so as to provide new ideas for the treatment of colon cancer.Methods:According to the exclusion criteria,patients with colon cancer were selected to collect cancer tissue samples,and normal volunteers were recruited to collect normal colonic mucosa tissue,Western blot was used to detect the expression of HDAC2 and acetylated p53k320 in colon cancer and normal colon mucosa;Chi square test was used to compare the expression of HDAC2 and acetylated p53k320 in different clinicopathological parameters;The correlation between HDAC2 and acetylated p53k320 was analyzed by person correlation analysis.Results:(1)The expression of HDAC2 was 1.6±0.04 in colon cancer and 0.29±0.06,compared with normal colon mucosa,the expression of HDAC2 in colon cancer tissue was up-regulated,and the difference was statistically significant(P<0.05);(2)The expression level of acetylated p53k320in colon cancer was 0.1±0.01 and 0.78±0.02 in normal colon mucosa,compared with normal colon mucosa,the expression of acetylated p53k320 was down regulated in colon cancer tissues,and the difference was statistically significant(P<0.05);(3)The expression of HDAC2 and acetylated p53k320 in colon cancer was related to the size of the tumor(P<0.05),the degree of differentiation(P<0.05),lymph node metastasis(P<0.05)and TNM stage(P<0.05);(4)There was a negative correlation between HDAC2 and acetylated p53k320in colon cancer tissues(P<0.05、r=-0.364).Conclusions:(1)Compared with normal colon mucosa,HDAC2 was highly expressed in colon cancer tissues,while acetylated p53k320 was low expressed in colon cancer tissues;(2)The larger the tumor,the lower the degree of differentiation,lymph node metastasis and TNM stage,the higher the expression of HDAC2 and the lower the expression of acetylated p53k320;(3)There was a negative correlation between HDAC2 and acetylated p53k320 in colon cancer tissues(P<0.05、r=-0.364).
