紫甘蓝对不同盐浓度处理的生理生化响应

【摘要】 福建位于沿海,土壤盐浓度较高,为确保紫甘蓝生产优质高产。本研究以紫甘蓝(Brassica oleracea L)品种‘紫甘85’为试验材料,采用水培方法,通过不同浓度NaCl处理,浓度分别为0(T1)、40(T2)、80(T3)、120(T4)、160(T5)nmol/L,研究不同浓度盐处理对紫甘蓝幼苗生长及生理响应、色素含量、光合作用、叶绿素荧光参数、叶片衰老和抗氧化酶系统的影响。主要研究结果如下:1、不同盐浓度处理对根系总长度、总表面积、体积、分根数、根尖数、根系直径、根系活力、株高、茎粗和干鲜重等均有不同程度的影响,其中T2(40nmol/L)处理有利于根系总长度、总表面积、体积、分根数、根尖数、根系平均直径和根系活力等的提高,超过一定盐浓度,随着盐浓度的不断提高,幼苗根系生长受到不同程度抑制;不同浓度盐处理均抑制幼苗株高的生长;T2处理有利于提高幼苗茎粗、干鲜重和根冠比,且T2-处理下的幼苗壮苗指数最大。说明T2处理最有利于紫甘蓝幼苗的生长。2、不同盐浓度处理对紫甘蓝幼苗色素含量、光合作用和叶绿素荧光参数的影响。结果表明,T2处理的紫甘蓝幼苗叶绿素a、叶绿素b和叶绿素(a+b)的含量均最高,类胡萝卜素含量则随盐浓度的提高而呈下降趋势;幼苗的净光合速率、蒸腾速率、胞间二氧化碳浓度、气孔导度和水分利用率均随着盐浓度的提高而下降;T2处理有助于提高幼苗的Fv/Fm、ΦPSⅡ和ETR,NPQ随着盐浓度的提高而提高,qP则随着盐浓度的提高而降低。3、不同盐浓度处理对紫甘蓝幼苗地上部和根系可溶性蛋白质、可溶性糖、丙二醛、脯氨酸含量和SOD、POD和CAT活性的影响。结果表明:盐处理有助于提高幼苗可溶性糖含量,且盐浓度越高,处理效果越明显;幼苗可溶性蛋白质含量则随着盐处理浓度的提高呈下降趋势,盐浓度越高,对可溶性蛋白质的抑制作用越明显;丙二醛和脯氨酸含量均随着盐处理浓度的提高而提高;SOD、POD和CAT的活性在处理5-15d时,随着盐处理浓度的提高而增加,处理15d后,T1和T2处理的幼苗SOD、POD和CAT活性呈上升趋势,而T3、T4和T5处理的幼苗SOD、POD和CAT活性则呈显著下降趋势,且浓度越高,下降趋势越明显。4、不同盐浓度处理紫甘蓝幼苗对定植后紫甘蓝鲜重、可溶性糖、可溶性蛋白质、维生素C、花青素和硝酸盐含量的影响。结果表明:T2和T3处理提高紫甘蓝鲜重,以T3处理对紫甘蓝鲜重促进作用最明显,T4和T5处理则降低紫甘蓝鲜重;T2、T3、T4和T5处理均提高了紫甘蓝可溶性糖含量,以T3处理效果最显著;可溶性蛋白质含量T2、T3和T4处理均有助于可溶性蛋白质含量的增加,以T3处理效果最明显,而T5处理则会降低可溶性蛋白质含量;T2、T3、T4和T5处理均有助于提高花青素含量,以T3处理下效果最明显;维生素C含量以T3处理效果最显著;T2处理下紫甘蓝硝酸盐含量最低,而T3、T4和T5处理的硝酸盐含量随着盐浓度提高而增加。通过盐浓度处理对紫甘蓝定植后产量及品质相关指标综合评价结果表明80nmol/L对幼苗处理最有利于产量及品质提高。
【Abstract】 Fujian is located in the coastal zone with high salt concentration in the soil to ensure the high quality and high yield of the purple cabbage production.In this study,the Brassica oleracea L variety "Purple Gan 85"was used as the test material,and 5 different concentrations of NaCl treatment were set up with hrdroponic method.The concentration was 0(T1),40(T2),80(T3),120(T4)and 160(T5)nmol/L respectively.The growth and physiological characteristics,pigment content and Photosynthesis of different concentration salt treatment on the seedlings of purple cabbage were studied.The effects of chlorophyll fluorescence parameters,leaf senescence and antioxidant enzyme system were studied.The main results are as follows:1、The results showed that different concentrations of salt treatments have different effects on total root length,total surface area,volume,root number,root tip number,root diameter,root activity,plant height,stem diameter and dry weight.The improvement of root length,total surface product,volume,root number,root tip number,root mean diameter and root vitality of root are most favorable for T2(40.00nmol/L)treatment.At a certain salt concentration,the root growth of seedlings was inhibited in varying degrees with the increase of salt concentration,and the growth of seedling height was inhibited by salt treatment at different concentrations.T2 treatment was beneficial to increase the stem diameter,dry weight and root shoot ratio of seedlings,and the maximum seedling index under T2 treatment was the largest.It indicates that T2 treatment is most conducive to the growth and development of purple cabbage seedlings.2、The effects of different concentrations of salt treatment on pigment content,photosynthesis and chlorophyll fluorescence parameters of purple cabbage seedlings were studied.The results showed that the contents of chlorophyll a,chlorophyll b and chlorophyll(a+b)in the seedlings treated with T2 were all the highest.The content of carotenoids decreased with the increase of salt concentration,and the net photosynthetic rate,transpiration rate,intercellular carbon dioxide concentration,stomatal conductance and water use rate decreased with the increase of salt concentration,and T2 treatment helped to improve the Fv/Fm of seedlings.PS,ETR and NPQ increase with the increase of salt concentration,while qP decreases with the increase of salt concentration.3、The effects of different salt treatment on the soluble protein,soluble sugar,malondialdehyde,proline content and SOD,POD and CAT activity in the shoot and root of purple cabbage were studied.The results showed that salt treatment could help to improve the soluble sugar content of seedlings,and the higher the concentration of salt,the more obvious the treatment effect.The content of soluble protein in the seedlings decreased with the increase of salt concentration,the higher the salt concentration,the more obvious the inhibition effect on soluble protein;the content of malondialdehyde and proline increased with the increase of salt concentration;SOD,POD and CAT.When the activity was treated with 5-15d,with the increase of salt concentration,the activity of SOD,POD and CAT in the seedlings treated with T1 and T2 increased,while the SOD,POD and activity of T3,T4 and T,decreased significantly,and the higher the concentration,the more obvious.4、The effects of different concentration salt treatment on the fresh weight,soluble sugar,soluble protein,vitamin C,anthocyanin and nitrate content of purple cabbage after colonization were studied.The results showed that T2 and T3 treatment improved the fresh weight of purple cabbage,and T3 treatment had the most obvious effect on the fresh weight of purple cabbage,T4 and T5 treatment reduced the fresh weight of purple cabbage.T2,T3,T4 and T5 treatment improved the soluble sugar content of purple cabbage,and the most significant effect was treated with T3;the soluble protein content T2,T3 and treatment were all helpful to the increase of soluble protein content.Obviously,T5 treatment could reduce the content of soluble protein;T2,T3,T4 and T5 could help to increase the content of anthocyanin,and the most obvious effect under T3 treatment;vitamin C content was the most significant with T3 treatment;the content of nitrate in purple cabbage under T2 treatment was the lowest,while T3,T4 and T5 treated nitrate content increased with the increase of salt concentration.The results of the comprehensive evaluation of the yield and quality of purple cabbage after the salt concentration treatment showed that the 80nmol/L was the most beneficial to the yield and quality of the seedlings.
