桃金娘枝叶中的间苯三酚类化学成分研究

【摘要】 桃金娘Rhodomyrtus tomentosa(Ait.)Hassk.为桃金娘科(Myrtaceae)桃金娘属(Rhodomyrtus)常绿灌木,又名岗稔、豆稔、乌肚子、山稔等,在我国广东、广西、云南等省区有广泛分布。桃金娘为岭南地区常用草药,其全株均可入药,且功能主治各不相同:叶主要用于治疗急慢性胃肠炎、痢疾等;根部多用于治疗肝炎、风湿性关节炎;而果实则具有养血止血、涩肠固精等功效。化学成分研究表明,桃金娘中主要含有三萜类、间苯三酚类、黄酮类、鞣质类及挥发油类成分,其中间苯三酚类化合物为其特征性成分之一。药理作用研究表明,部分间苯三酚类化合物显示出较好的抗菌、抗病毒、抗疟、抗肿瘤等活性。本课题组前期对多种桃金娘科药用植物中的间苯三酚类成分进行了系统的化学成分研究,并已取得了一系列的新发现。目前,文献报道仅从桃金娘中分离得到了10个间苯三酚类化合物,其研究尚不充分。为进一步发现结构新颖、活性良好的间苯三酚类化合物,本论文对桃金娘枝叶中的该类成分开展了系统研究。运用多种色谱分离技术,从桃金娘枝叶95%乙醇提取物的石油醚萃取部位分离得到了20个间苯三酚类化合物,并通过NMR、单晶X衍射及计算化学等方法确定了它们的结构,分别为:tomentone A(RT-1)、tomentone B(RT-2)、tomentone C(RT-3)、tomentone D(RT-4)、rhodomyrtosone R(RT-5)、rhodomyrtosone U(RT-6)、rhodomyrtosone V(RT-7)、rhodomyrtosone C(RT-8)、tomentosone A(RT-9)、rhodomyrtone(RT-10)、rhodomyrtosone B(RT-11)、rhodomyrtosone J(RT-12)、rhodomyrtosone M(RT-13)、rhodomyrtosone O(RT-14)、rhodomyrtosone S(RT-15)、rhodomyrtosone I(RT-16)、rhodomyrtosone K(RT-17)、rhodomyrtosone N(RT-18)、rhodomyrtosone P(RT-19)和endoperoxide G3(RT-20)。上述化合物中,RT-1~RT-4为4个具有新颖骨架结构的环聚酮-倍半萜杂合物,RT-5~RT-7为3个新的环聚酮-间苯三酚-环聚酮三聚体,RT-12~RT-15和RT-17~RT-19为7个新的环聚酮-间苯三酚二聚体。

【Abstract】 The plant Rhodomyrtus tomentosa(Ait.)Hassk.is an evergreen shrub of family Myrtaceae and widely distributed in south China,including Guangdong,Guangxi,Yunnan and Fujian provinces.The whole plant of R.tomentosa was used as folk medicine for the treatment of colic diarrhea,dysentery,hemorrhage,abscesses,rheumatic arthritis and hepatitis.A series of triterpenes,phloroglucinols,flavonoids,tannins and essential oil have been isolated from this plant.Phloroglucinols are one of the characteristic components of R.tomentosa,some of which showed potent antibacterial,antimalarial and anticancer activities.To date,however,only 10 phloroglucinols have been isolated from R.tomentosa.As a part of our onging program to search for structurally unique and biologically interesting phloroglucinol derivatives,we carried out systematically phytochemical investigation on the twigs and leaves of R.tomentosa,which led to the isolation of 20 phloroglucinol derivatives.Their structures were established based on extensive spectral methods.They are tomentone A(RT-1),tomentone B(RT-2),tomentone C(RT-3),tomentone D(RT-4),rhodomyrtosone R(RT-5),rhodomyrtosone U(RT-6),rhodomyrtosone V(RT-7),rhodomyrtosone C(RT-8),tomentosone A(RT-9),rhodomyrtone(RT-10),rhodomyrtosone B(RT-11),rhodomyrtosone J(RT-12),rhodomyrtosone M(RT-13),rhodomyrtosone O(RT-14),rhodomyrtosone S(RT-15),rhodomyrtosone I(RT-16),rhodomyrtosone K(RT-17),rhodomyrtosone N(RT-18),rhodomyrtosone P(RT-19)and endoperoxide G3(RT-20).Among them,RT-1~RT-7,RT-12~RT-15 and RT-17~RT-19 are 14 new phloroglucinol derivatives.Especially,RT-1~RT-4 are four new phloroglucinol-terpene adducts possessing a novel skeletal type.
