打破紫斑牡丹种子休眠因素及抗氧化酶活性变化的研究

【摘要】 紫斑牡丹为芍药科芍药属的国家三级保护植物。在药用、观赏、油用方面具有很高的价值。但由于紫斑牡丹种子休眠的特性,使其种子繁殖具有萌发历时长,出苗不齐,萌发系数低等问题。因此,本试验以紫斑牡丹种子为材料,研究打破其下胚轴和上胚轴休眠的最适方法还有种子萌发过程中抗氧化酶活性的变化,为生产上快速培育紫斑牡丹优质种苗提供技术支持。结果表明:不同处理对紫斑牡丹种子生根影响不同。浓硫酸3min处理的种子,生根率为38.9%。光照强度为3000Lx,处理时间为16h时,最适于其生根,生根率为89.3%;当温度为25℃、处理时间为24h时,生根率为90.9%;GA3处理可以加快紫斑牡丹种子生根速度、提高生根率。其中,300 mg ·L-1GA3处理时间为24h最有利于紫斑牡丹种子生根,生根率最高,为93.7%。2.不同处理对紫斑牡丹种子发芽的影响不同。层积处理30d时发芽率为93.4%,发芽势为82.9%;300 mg·L-1GA3溶液、处理时间为24h时,其发芽率为93.2%,发芽势为89.4%;用100 mg ·L-1乙烯处理的种子24h,其发芽率为89.8%,发芽势为89.1%。3.在单因素试验基础上,采用L25(56)正交试验法,研究了不同层积时间、不同GA3浓度处理及不同处理时间、不同乙烯浓度处理及不同处理时间。其中,层积30d、GA3浓度为300mg·L-1、处理时间为12h、乙烯浓度为200 mg·L-1、处理时间为24h最有利紫斑牡丹种子发芽,发芽率为95.2%。4.在紫斑牡丹种子萌发过程中,抗氧化酶活性的变化规律不同,超氧化物歧化酶(SOD)、过氧化物酶(POD)在紫斑牡丹种子萌发过程中,呈下降趋势,过氧化氢酶(CAT)呈逐渐上升趋势。

【Abstract】 Paeonia rockii is a national grade 3 protected plant of paeonia lactiflora.It has high value in medicine,ornamental and oil use.However,due to the dormancy of the seeds of Paeonia rockii,the seed propagation has such problems as long germination duration,uneven seedling emergence and low germination coefficient.Therefore,this experiment took the seeds of Paeonia rockii as materials to study the optimal method to break the dormancy of its hypocotyl and epicotyl,as well as the change of antioxidant enzyme activity during seed germination,so as to provide technical supportfor the rapid cultivation of high-quality seedlings of Paeonia rockii.The results show that:1.The rooting effects of different treatments were different.The rooting rate of seeds treated with concentrated sulfuric acid for 3min was 38.9%.When the light intensity was 3000Lx and the processing time was 16h,the rooting rate was 89.3%.When the temperature is 25℃,the processing time for 24 h,the rooting rate is highest,at 90.9%.GA3 treatment can accelerate the rooting speed and improve rooting rate.Among them,300 mg·1-1GA3 treatment time of 24h was the most favorable for the rooting of peony puiple-spot seeds,and the rooting rate was 93.7%.2.Different treatments had different effects on seed germination of Paeonia rockii.The germination rate and germination potential were 93.4%and 82.9%at 30d.The germination rate and germination potential of 300 mg·1-1GA3 solution for 24h were 93.2%and 89.4%respectively.The seed treated with 100 mg·1-1 ethylene for 24 h had a germination rate of 89.8%and a germination potential of 89.1%.o3.On the basis of single factor test,L25(56)orthogonal test method was used to study different lamination time,different concentration of GA3 and different treatment time,different concentration of ethrel and different treatment time.Among them,the concentration of laminate 30d and GA3 was 300mg·1-1,the treatment time was 12h,the concentration of ethrel was 200mg·1-1,and the treatment time was 24h,germination rate is 95.2%.4.During the seed germination of Paeonia rockii.,the activity of antioxidant enzymes showed different dynamic patterns.During the seed germination of Paeonia rockii.,superoxide gasification enzyme(SOD)and peroxidase(POD)showed a decreasing trend,while catalase(CAT)showed a gradually increasing trend.
