环状RNA circHIPK3在肺腺癌中的表达及临床意义

【摘要】 背景与目的:环状RNA(circular RNA,circRNA)是一种共价闭合环形RNA分子,广泛存在于各种生物细胞中,参与基因的表达调控,具有高度保守性、组织发育特异性和稳定性,在疾病发生过程中存在异常表达,具有潜在的诊断价值。在肺癌组织中,已发现相关circRNA通过内源性调控作用参与肺癌的发生发展,差异表达的circRNA与肺癌的临床特征存在相关性,有可能成为肺癌新的诊断标记物。circHIPK3是一种丰度很高的环状RNA,具有一定的稳定性,在多种肿瘤中存在差异表达,并参与肿瘤的发生发展。进一步发掘其在肺腺癌组织中的差异表达情况,为肺腺癌的诊断提供依据。方法:本研究共收集54例配对的肺腺癌组织及癌旁组织标本,并收集了患者的临床病理资料。采用实时荧光定量聚合酶反应(quantitative real time polymerase chain reaction,qRT-PCR)方法检测circHIPK3在54例肺癌组织与癌旁组织中的表达水平,使用配对t检验分析circHIPK3在肺腺癌组织与癌旁组织中表达水平的差异。采用独立样本t检验分析肺腺癌患者circHIPK3表达水平与临床病理特征之间的关系,并使用受试者工作特征曲线来评估circHIPK3对肺腺癌组织的诊断价值。结果:(1)环状RNA circHIPK3表达于肺腺癌组织及癌旁组织中。(2)肺腺癌组织中circHIPK3的表达水平低于癌旁组织(P<0.01)。(3)环状RNA circHIPK3的表达水平与肺腺癌的TNM分期(P=0.005)、淋巴结转移(P=0.007)、肿瘤最大径(P=0.025)存在明显相关性。(4)环状RNA circHIPK3对肺腺癌组织具有诊断价值,其ROC曲线下的面积是0.831(P<0.01),敏感度为64.8%,特异度为88.9%。结论:环状RNA circHIPK3在肺腺癌组织中表达下调,与肺腺癌的病理特征存在一定的相关性,对肺腺癌组织的诊断具有一定价值。

【Abstract】 Objective: Being covalently closed circular RNA molecules,circular RNAs(circRNAs)are widely present in a variety of biological cells and involve in gene expression regulation,with a high degree of conservation and stability.In addition,they exhibit tissue-and developmental stage-specific expression.Circular RNAs are often abnormally expressed during the pathogenesis of diseases and thus are of great diagnostic value.In lung cancers,circRNAs participate in the development and progression of lung cancers through endogenous regulation.Given that differentially expressed circRNAs are correlated with the clinical features of lung cancers,these circRNAs may serve as diagnostic markers for lung cancers.circHIPK3 is an abundant and stable circRNA expressed in various tumors,which participate in the tumor development and progression.Further investigation of the differential expression of circHIPK3 in lung adenocarcinoma is conducive to the development of novel approaches for diagnosis of lung adenocarcinoma.Methods: The 54 paired lung adenocarcinoma tissues and adjacent nontumorous tissues were collected from surgical lung cancer patients,meanwhile the clinical pathological data of the 54 patients were collected.Expression level of circHIPK3 was detected in lung adenocarcinoma and paired adjacent nontumorous tissues by quantitative real time polymerase chain reaction(qRT-PCR),differences in expression level of circHIPK3 were analyzed by the paired t-test.The relationships between circHIPK3 expression levels in lung adenocarcinoma tissues and clinicopathological features were analyzed by two sample t-test,a receiver operating characteristic(ROC)curve was established to estimate the diagnostic value of circHIPK3 in lung adenocarcinoma.Results:(1)circHIPK3 was naturally expressed in lung adenocarcinoma tissues.(2)The expression levels of circHIPK3 were lower in lung adenocarcinoma tissues than paired adjacent nontumorous tissues(P<0.01).(3)The expression levels of circHIPK3 were associated with the TNM stage(P =0.005),lymphatic metastasis(P =0.007)and tumor size(P=0.025).(4)The expression levels of circHIPK3 have diagnostic values for lung adenocarcinoma tissues.The area under the ROC curve was 0.831(P<0.01),the sensitivity was 64.8%,the specificity was 88.9%.Conclusion:circHIPK3 was downregulated in lung adenocarcinoma tissues.The expression levels of circHIPK3 have a correlation with the pathological features of lung adenocarcinoma.circHIPK3 has certain value for the diagnosis of lung adenocarcinoma.
